摘要
目的:研究miR-300对人卵巢癌细胞增殖、凋亡和周期的影响。方法:通过荧光实时定量PCR(qRT-PCR)检测卵巢正常上皮细胞及卵巢癌细胞中miR-300的表达水平和miR-300 siRNA的沉默效果;CCK-8法检测沉默miR-300对卵巢癌细胞增殖的影响;流式细胞术检测沉默miR-300对卵巢癌细胞凋亡和周期的影响。结果:miR-300在卵巢癌细胞中的表达显著高于卵巢正常上皮细胞;miR-300 siRNA可有效沉默卵巢癌细胞中miR-300的表达;沉默miR-300的表达可以明显抑制卵巢癌细胞的增殖能力,增强卵巢癌细胞的凋亡能力,并使卵巢癌细胞的周期阻滞于G_1期。结论:miR-300在卵巢癌细胞的增殖、凋亡和周期过程中起着重要的调控作用。
Objective: To investigate the effects of miR- 300 on the proliferation,apoptosis,and cell cycle of humanovarian cancer cells. Methods: The expression of miR- 300 in the normal ovarian epithelial cell and ovarian cancer cells,and the silencing efficiency of miR- 300 siRNA was detected by quantitative Real time PCR( qRT-PCR). After siRNA and NC transfected into ovarian cancer cells for 48 hours,the proliferation of ovarian cancer cells were analyzed by CCK- 8,the apoptosis and cell cycle of ovarian cancer cells was analyzed by flow cytometry. Results: The expression of miR- 300 was significantly upregulated in ovarian cancer cells comparing with normal ovarian epithelial cells and could be effectively silenced by miR- 300 siRNA. Proliferation of ovarian cancer cells were inhibited after transfected miR- 300 siRNA,but cells apoptosis were promoted. Cell cycle of ovarian cancer cells were arrested at stage of G_1 after transfected miR- 300 siRNA. Conclusion: miR- 300 play an important role on the proliferation,apoptosis and cell cycle of ovarian cancer cells.
