摘要
目的:建立Lewis肺癌荷瘤小鼠动物模型,研究肺癌细胞中PTHrP、MCT1表达的相关性及与癌细胞衰老的关系和意义。方法:C57BL/6小鼠共30只,随机分三组:正常小鼠组(第1组)、早期肿瘤组(第2组)、晚期肿瘤组(第3组)。免疫组织化学法检测三组中PTHrP、MCT1的表达。结果:三组动物细胞PTHrP蛋白的分泌及MCT1的阳性表达率高度一致,且与衰老相关指标βGal的表达成明显负相关性;两个指标在第3组的表达较在第2组和第1组中的表达明显升高,结果有统计学意义(P<0.05);在第2组的表达较在第1组中的表达明显升高,结果有统计学意义(P<0.05)。结论:晚期肿瘤组织PTHrP、MCT1的表达具有显著正相关性,且与衰老相关指标βGal的表达成明显负相关性。
Objective: To establish Lewis lung cancer bearing mice animal model,and study the correlation between MCT1 and PTHrP expression in lung cancer cells and the relationship with cancer cell senescence. Methods: A total of 30 C57 BL /6 mice were randomly divided into three groups: normal mice group( first groups),early cancer group( second group),and advanced cancer group( third groups). The expression of PTHrP and MCT1 was detected by immunohistochemistry. Results: The positive expression of three groups of animal cells PTHrP protein secretion and MCT1 rate highly consistent,and significantly negatively correlated with senescence associated markers of beta Gal expression,two indicators expression in group 3 than in group 2 and group 1 significantly( P〈0. 05). In group 2 expression than in group 1 expression was significantly increased( P〈0. 05). Conclusion: The expression of PTHrP and MCT1 in the advanced tumor tissues had significant positive correlation,and the expression of beta Gal was negatively correlated with the aging related indexes.
