Expression and clinical significance of FGFR1 and MMP3 in esophageal squamous cell carcinoma

摘要 目的:观察成纤维细胞成长因子受体1(fibroblast growth factor receptor 1,FGFR1)和基质金属蛋白酶3(matrix metalloproteinase 3,MMP3)基因和蛋白在食管鳞癌、癌旁及正常食管组织中的表达情况,探讨它们与食管鳞癌患者临床病理特征之间的关系。方法:采用RT-q PCR、Western blot和免疫组织化学染色检测78例食管鳞癌组织(鳞癌组)、35例食管鳞癌癌旁组织(癌旁组)及35例正常食管组织(正常组)中FGFR1和MMP3 mRNA及蛋白的表达,并分析其与肿瘤临床病理特征之间的关系。结果:FGFR1和MMP3 mRNA在食管鳞癌组表达水平显著高于癌旁组和正常组,差异具有统计学意义(P<0.05)。在癌旁组和正常组表达差异无统计学意义(P>0.05)。FGFR1和MMP3蛋白在食管鳞癌组中的表达率和表达水平明显高于癌旁组和正常组,差异具有统计学意义(P<0.05)。在癌旁组及正常组表达率及表达水平差异均无统计学意义(P>0.05)。FGFR1和MMP3蛋白在食管鳞癌中的表达与患者性别、年龄、肿瘤分化程度无关(P>0.05),而与肿瘤浸润深度、淋巴结是否转移及TNM分期相关(P<0.05)。食管鳞癌中,FGFR1和MMP3的蛋白表达呈正相关(r=0.303,P<0.05)。结论:FGFR1和MMP3在食管鳞癌中均高表达,两者可能与食管癌的侵袭和转移有关。 Objective:To investigate the expression of fibroblast growth factor receptor 1 (FGFR1) and matrix metalloproteinase 3 ( MMP3 ) in esophageal squamous cell carcinoma tissues, para - tumor tissues and normal esopha- geal tissues, and the association with clinical pathological features. Methods: Real - time qPCR, immunohistochemis- try and Western blot were used to detect the expressions of FGFR1 and MMP3 mRNA and protein levels in esophageal squamous cell carcinoma tissues ( 78 cases), the corresponding para - tumor tissues ( 35 cases ) and normal esophage- al tissues (35 cases). Their relationships with clinical pathological characteristics of esophageal squamous eel1 carci- noma were analyzed. Results: The expression of FGFR1 and MMP3 mRNA in esophageal squamous cell carcinoma tis- sues were much higher than those in para - tumor tissues and normol esophageal tissues ( P 〈 0.05 ). No significant difference was observed between para - tumor tissues and normol esophageal tissues ( P 〉 0.05 ). The positive rate and expression level of FGFR1 and MMP3 protein in esophageal squamous cell carcinoma tissues were significantly higher than those in para - tumor tissues and normal esophageal tissues ( P 〈 0.05 ). No significant difference was ob- served between para- tumor tissues and normol esophageal tissues (P 〉 0.05 ).The expression of FGFR1 and MMP3 protein in esophageal squamous cell carcinoma was not associated with gender, age, differentiation of carcinoma ( P 〉 0.05 ), but associated with the depth of invasion, lymphatic metastasis and tumor TNM stages ( P 〈 0.05 ). The ex- pression level of FGFR1 in esophageal squamous cell carcinoma was positively correlated with the expression of MMP3 (r = 0.303,P 〈 0.05 ). Conclusion:The expression of FGFR1 and MMP3 are significantly higher than those in para -tumor tissues and normal esophageal tissues. They may be associated with invasion and metastasis of esophageal squamous carcinoma.
