摘要
目的研究瞬时受体电位香草醛亚家族1(TRPV1)在高脂处理肝细胞中的变化及其作用。方法将肝细胞(BRL-3A)分为对照组和250、500μmol·L^(-1)软脂酸钠组,加入辣椒素和辣椒卓平对肝细胞进行干预,通过Q-PCR法测量肝细胞中TRPV1的表达水平,并采用流式细胞仪检测肝细胞的凋亡水平。结果肝细胞的凋亡比例随软脂酸钠浓度的增加而增加,其浓度为500μmol·L^(-1)时,TRPV1的表达水平明显增加;辣椒素能够上调TRPV1的表达水平并减轻高脂所诱导的肝细胞凋亡;而辣椒卓平能够逆转辣椒素对肝细胞的保护作用。结论高脂能够激活肝细胞中的TRPV1通道,同时,上调TRPV1,减少高脂诱导的肝细胞凋亡。
OBJECTIVE To investigate the alteration of transient receptor potential vanilloid 1 ( TRPV1 ) in hepatocytes treated with high fat and its role in this process. METHODS The hepatic cells (BRL -3A) were divided into three groups:control group,250 μmol·L^-1 triglyceride group and 500 μmol·L^-1 triglyceride group. Then, capsaicin and capsazepin were administrated into hepatocytes for further intervention. The expressions of TRPV1 and hepatocytes apoptosis were measured by Q - PCR and flow cytometer respectively. RESULTS The apoptotic ratio of hepatocytes was concentration - dependent to high fat and the TRPV1 mRNA expression was significantly increased at 500 μmol·L^-1 triglyceride. Moreover, capsaicin obviously reduced the apoptosis of hepatocytes induced by high fat via increasing TRPV1 expression ,which was reversed by the application of capsazepin. CONCLUSION High fat can activate TRPV1 pathway in hepatocytes, and TRPV1 enforcement is able to alleviate the hepatocytes apoptosis.
