阿达木单抗治疗恒河猴胶原性关节炎的药效学研究

Pharmacodynamic study of Adalimumab on the rhesus monkey arthritis induced by collagen

目的研究恒河猴胶原性关节炎(CIA)模型用于治疗类风湿性关节炎的药物评价。方法恒河猴被胶原免疫后,给予阿达木单抗1.0 mg·kg-1和阿达木单抗(0.5 mg·kg-1)联合甲氨蝶呤(1.0 mg·kg-1)进行治疗,观察给药前、后猴的临床表现并进行临床评分,检测各项炎症指标,并进行四肢关节的X光影像学分析。结果免疫后,猴有不同程度的类风湿性关节炎症状出现;给药后,猴的受累关节病情好转或未持续恶化。结论建立的CIA恒河猴模型的病程稳定,可用于类风湿性药物的药效评价。

OBJECTIVE To study the feasibility of using collagen - induced arthritis （CIA） model in rhesus monkeys to further evaluate the efficacy of drugs treating of rheumatoid arthritis （RA）. METHODS Rhesus monkeys were subcutaneously injected with bovine type II collagen for immunization. The monkeys were treated with Adalimumab （0.5 mg·kg^-1） or Adalimumab （0.5 mg·kg^-1） combined with Methotrexate （ 1.0 mg·kg^-1） after the immunization. The clinical manifestations were observed, and clinical scores were performed before and after the administration, and the inflammation related indicators were also detected. The X - rays of joints were analysized. RESULTS Arthritis symptoms were found in the monkeys after the immunization. The inflammation was controlled after the treatment,and the joints were recovered or stopped worsening. CONCLUSION The CIA rhesus monkey model is stable enough and can be used to evaluate the effect of RA treatment.
