摘要
为探讨质外体抗冻蛋白能否作为冬油菜抗寒评价的一项新指标,本研究利用SDS-PAGE凝胶电泳对白菜型冬油菜‘陇油7号’和‘陇油9号’与优质春油菜Parkland及其杂交F1代质外体抗冻蛋白进行了检测。结果表明,低温胁迫前所有单株质外体抗冻蛋白电泳图谱颜色浅,其含量较低。低温胁迫后,各单株质外体抗冻蛋白电泳图谱颜色加深,含量增加,且单株间差异较大。冬性亲本和能够越冬单株质外体抗冻蛋白电泳图谱颜色深,而春性亲本与不能越冬单株的颜色浅。由此说明,质外体抗冻蛋白作为抗寒评价的新指标是可行的,试验中筛选出的一些抗寒性状优良的单株,可为冬油菜抗寒品种的选育和改良提供一定的基础材料和理论依据。
In order to explore a new evaluation of cold resistance and the basis for equality improvement and cold resistance breeding in winter turnip rape (Brassica rapa L. ) in the north of China, the apoplast antifreeze proteins in winter turnip rape Longyou7, Longyou9, spring turnip rape Parkland and their first-filial generation were detected by SDS-PAGE. The result showed that the SDS-PAGE electrophoretogram of the apoplastic antifreeze protein in individual plant were lighter before low temperature stress, and its content decrease; And darker after low temperature stress, and its content increased. The SDS-PAGE eleetrophoretogram of the apoplastic antifreeze protein in winter parents and the individuals which could survive in winter was darker than that of the spring parent and the individuals which could not survive in winter based on the individual overwintering. It was feasible to evaluate cold resistance by apoplast antifreeze protein as new index, and some excellent cold resistance plants were selected, which could provide the basis for equality improvement and cold resistance breeding in winter turnip rape.
