摘要
以丽格海棠品种巴克斯(Barkos)的叶片为外植体,研究光照强度和时间、植物生长调节剂对不定芽诱导的影响;植物生长调节剂对不定芽增殖的影响;组培苗生根的最佳培养基。结果表明:丽格海棠叶片的最佳不定芽再生培养基为MS+1mg/L TDZ+0.1mg/L 2,4-D或MS+1mg/L 6-BA+0.1mg/L TDZ,给予40d弱光培养可促进不定芽再生,再生率可达88.3%;不定芽最佳增殖培养基为2mg/L 6-BA+0~0.1mg/L NAA,组培苗最佳生根培养基为MS+0.1~0.3mg/L IBA。
In this experiment, the explants is Rieger Bego (Barkos) of leaves, we study the light intensity and plant growth regulator on influence of the adventitious bud induction; Plant growth regulator effects on adventitious bud proliferation and rooting of tissue culture seedling. The results show that the Rieger Bego optimum adventitious bud of leaves regeneration medium for MS + 1 mg/L TDZ + 0.1 mg/L 2, 4-D or MS + 1 mg/L 6-BA + 0.1 mg/L TDZ, weak light training can promote in vitro shoot regeneration in 40d, regeneration rate is 88.3 %; Best adventitious bud proliferation culture medium for 2 mg/L 6-BA + 0-0.1 mg/L NAA; The best rooting culture medium for MS + 0.1-0.3 mg/L IBA.
