摘要
目的:研究黄精多糖(polygona-polysaccharose,PSP)对糖尿病大鼠模型的保护作用机制。方法:将60只健康SD大鼠随机分成5组,空白对照组、模型组、黄精多糖低、中、高剂量组。应用链脲佐菌素(streptozotocin,STZ)造模成功后,黄精多糖低、中、高剂量组给予低(200 mg/kg)﹑中(400 mg/kg)﹑高(800 mg/kg)剂量黄精多糖溶液灌胃干预12周。每周称量大鼠空腹体质量;造模成功后2、4、6、8、10、12周测量各组大鼠基础进食量﹑饮水量﹑尿量。分别于造模前3 d、造模后3 d、造模成功后每两周采用葡萄糖氧化酶法测空腹血糖值(fasting blood glcose,FBG);第4、8、12周末心脏取血测量糖化血红蛋白(glycosylated hemoglobin,Hb Alc)﹑C肽(C-peptide,C-P)﹑胰岛素(insulin,INS)、血清超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)值;于造模成功后4、8、12周处死大鼠取胰腺行HE染色病理组织学观察。结果:模型组进食量、饮水量、尿量、空腹血糖、糖化血红蛋白、MDA水平均明显高于空白对照组(P<0.05),基础体质量、血清胰岛素、C肽含量、血清SOD均明显低于空白对照组(P<0.05)。黄精多糖低、中、高剂量组进食量、饮水量、尿量、血糖水平、糖化血红蛋白、MDA水平均高于空白对照组(P<0.05),基础体质量、血清胰岛素、C肽含量、血清SOD均低于空白对照组(P<0.05)。与模型组比较,黄精多糖中、高剂量组胰腺组织明显形态完善,胰岛细胞明显增多。结论:黄精多糖能改善STZ诱导的糖尿病大鼠症状,显著降低血糖、血清糖化血红蛋白浓度,明显提高组织胰岛素及C-肽表达量、增强胰岛素敏感性,改善糖尿病大鼠胰岛素抵抗。
Objective: To research the protective mechanism of polygona-polysaccharose on rat model of di- abetes. Methods: The 60 healthy SD rats were randomly divided into 5 groups, blank control (BC) group, dia- betic model(DM) group and other groups as different doses of drug intervention group after they were modelled. The rats lived in clean laboratory and free drinking water, eating, and adaptive feeding a week. With 3 different concentrations of polygona-polysaccharose solution [low (200 mg/kg), middle (400 mg/kg), high (800 mg/kg)] to in- tervent the diabetic rats and lasted 12 weeks. And we test glycosylated hemoglobin (HbAlc), C peptide(C-P), in- sulin(INS), serum SOD, MDA indicators by heart punctures on 3 days before making model, 3 days after making model and 4th, 8th,12th week; Weighing rats weight on an empty stomach every week, and measurement fasting blood glucose (glucose oxidase method) after vein blood; Test basic food intake, water intake, urine volume after the success of modeling 2,4,6,8,10,12 weeks; After the success in modeling the 4, 8, 12 weeks the rats were killed and we observed the pancreas after HE staining. Results: Compared with the rats in BC group, the rats in DM group showed that food intake, water intake, urine volume, blood glucose levels, glyeosylated hemoglobin, serum SOD, MDA levels were increased significantly (P〈0.05); But the basis weight, serum insulin, C peptide levels were significantly lower (P〈0.05). Compared with the rats in BC group, the rats in L, M, H groups showed that food intake, water intake, urine volume, blood glucose levels, glycosylated hemoglobin, serum SOD, MDA levels increased (P〈0.05) and basis weight, serum insulin, C peptide levels were lower (P〈 0.05); Compared with the rats in model group, the morphology of pancreatic tissue in M and H groups im- proved and the islet cells were significantly increased. Conclusion: Polygona-polysaccharose can improve the clinical symptoms of diabetic rats induced by STZ, it can significantly reduce the blood sugar, blood lipid lev- els and the concentration of glycated hemoglobin, and significantly improve the expression of insulin and C- peptide, enhance sensitivity of the insulin, improve lipid metabolic disturbance and insulin resistance in diabetic rats.
