不同炮制时间制何首乌大鼠血清中大黄素含量测定 被引量:4

Determination of Emodin Content in Serum of Rats in Different Processing Time of Zhi Heshouwu(Polygonum Multiflorum)

摘要 目的:研究不同炮制时间制何首乌经口服给药后大鼠血清中大黄素含量的变化。方法:将20只SD大鼠随机分成空白组、制何首乌不同炮制时间24、48、72 h组,每组5只,共4组。空白组给生理盐水。其余各组分别给予不同炮制时间的制何首乌水提液灌胃,24 h(12 g/kg)、48 h(12 g/kg)、72 h(12 g/kg),连续7 d后,腹主动脉取血,离心,采用高效液相仪分析大鼠血清中大黄素的含量。HPLC条件,色谱柱:Spursil ODS 5μm 250×4.6 mm色;流动相甲醇∶磷酸水(83∶17);紫外检测波长:254 nm。结果:制何首乌炮制24 h时大鼠血清中大黄素含量为0.01536μg/L,炮制48 h血清含量为0.01276μg/L,炮制72 h血清含量为0.0100μg/L。结论:制何首乌不同炮制时间给大鼠经口服给药后,血清中大黄素的含量会发生改变,且随炮制时间的延长,大鼠血清中的大黄素含量呈减少趋势。提示在使用何首乌时,可以通过延长其炮制时间,减少何首乌中大黄素的含量,可能会降低其对肝脏的损伤。 Objective: To study the changes of serum emodin in Polygonum different processing time after o- ral administration. Methods: The 20 SD rats were randomLy divided into blank group, Polygonum different pro- cessing times (24 h, 48 h, 72 h). Each groups is five, a total of four groups. Blank group was given saline. Other groups were given different processing time water extract of Polygonum [24 h(12 g/kg), 48 h(12 g/kg), 72 h(12 g/kg)]. After continuous 7 d, abdominal aortic blood was taken, centrifugal. HPLC was used to analyze the emodin in rat serum. HPLC conditions: Column Spursil ODS 5 μm 250×4.6 mm column; mobile phase methanol: aqueous phos- phoric acid (83:17); detection wavelength 254 nm. Results: Processing 24h of Polygonum multiflorum Thunb in rat serum content of emodin 0.01536 μg/L, processing 48 h in serum 0.01276 μg/L, processing 72 h serum content 0.0100 μg/L. Conclusion: Radix Polygoni Mnltiflori from different processing time to rats by oral administration, the content of emodin in serum will change and emodin content in serum of rats was decreased with the exten- sion of processing time. It is revealed that when using the Polygonum, it can prolong the processing time, reduce the content of emodin in Radix Polygoni Muhiflori, which may reduce the damage to the liver.
