益智宁对注意缺陷多动障碍动物模型SHR大鼠学习记忆的影响 被引量:1

摘要 目的:探讨益智宁对注意缺陷多动障碍动物模型SHR大鼠学习记忆能力的影响。方法:将50只幼年SHR大鼠随机分为益智宁高、中、低剂量组、空白对照组、哌甲酯对照组,每组10只,益智宁高、中、低剂量组分别给予益智宁煎药液(14.4 g/kg,7.2 g/kg,3.6 g/kg),哌甲酯对照组给予哌甲酯(0.0015 g/kg),空白对照组给予生理盐水,每组均按12.5 m L/kg等容量灌胃给药。给药4周后,通过Morris水迷宫检测SHR大鼠学习记忆能力,定位航行实验5 d,空间探索实验1 d。结果:在定位航行实验中,空白对照组的逃避潜伏期表现不稳定,各用药组潜伏期随着训练时间的增加,均有逐渐缩短趋势,到训练第5天,各组间比较,差异有统计学意义(P<0.05);益智宁高剂量组、哌甲酯对照组逃避潜伏期明显缩短,与空白对照组比较,差异有统计学意义(P<0.05),但益智宁高剂量组与哌甲酯对照组比较,差异无统计学意义(P>0.05);在空间探索实验中,益智宁高剂量组在目标象限游泳的路程最长,与空白对照组比较,差异有统计学意义(P<0.05),哌甲酯对照组及益智宁高剂量组在目标象限游泳的时间及穿台次数均高于空白对照组,差异有统计学意义(P<0.05),但哌甲酯对照组与益智宁高剂量组比较,差异无统计学意义(P>0.05)。结论:益智宁高剂量及哌甲酯均能改善SHR大鼠的学习能力和记忆能力。 Objective: To explore the effect of Yizhining decoction on the working memory of Spontaneously Hyperactivity Rat (SHR)-An animal model of Attention Deficit Hyperactivity Disorder. Methods: The 50 SHR were randomly divided into Yizhining high dose group, Yizhining medium dose group, Yizhining low dose group, Methylphenidate Hydrochloride group (MH) and Blank control group ,with 10 rats in each group . The Yizhining group were treated by Yizhining decoction(14.4 g/kg, 7.2 g/kg, 3.6 g/kg); the MH group was applied with Ritalin (0.0015 g/kg), while the blank control group given normal saline .The gastric volume to the stomach was equal (12.5 mL/kg), once a day for 4 weeks. After 4 weeks, Morris Water Maze test, which covered orienting naviga- tion test for 5 consecutive days and one day's spatial probe test, was performed to investigate the working memory of each group. Results: In the orienting navigation test, the escape latency of the blank control group fluctuated while the other groups declined with the increase training days .When it comes to the fifth day, there was significant difference within them (P〈0.05). Compared with blank control group, the escape latency of the Yizhining high dose group and the MH group was significantly shorter (P〈0.05), while there was no notable dif- ference between these two groups (P〉0.05). In the spatial probe test, the high dose of Yizhining spent more time in the target quadrant, and the difference was outstanding compared with the blank control group (P〈0.05). The times of platform-crossing of the Yizhining high dose group and the MH group were more than the other three groups, as well as the time rats spent in the target quadrant. And the difference was remarkable compared withthe blank control group (P〈0.05), while there was no significant difference between the Yizhining high dosegroup and the MH group (P〉0.05). Conclusion: The high dose of Yizhining decoction can improve the working memory of SttR as well as the Methylphenidate Hydrochloride.
