摘要
设计3个冷藏组(7、14、21 d)和2个对照组(未处理、常温21 d),通过厌氧发酵接种物的理化指标(p H值、VFA、TIC、NH4+-N)测定、菌落结构分析和厌氧发酵产甲烷能力测试,研究了4℃冷藏处理对厌氧发酵接种物活性的影响。结果表明:接种物在4℃冷藏过程中,代表产甲烷菌的优势条带随冷藏时间的延长出现了亮度下降,并有少量条带丢失的现象;其厌氧发酵产甲烷能力出现下降,前15 d的日产甲烷速率明显低于未处理组的;但冷藏处理能够减缓氨氮浓度的增高,有助于接种物的厌氧发酵活性的恢复。
In this study, the inoculum for anaerobic fermentation was stored under ilve different conditions formed by 3 cold (4 ℃ ) storage groups (7 d, 14 d, 21 d) and 2 control groups (untreated storage, room-temperature storage for 21 d), its physical and chemical properties (pH-value, VFA, TIC, and NH4+-N), colony structure and anaerobic methanogenesis ability were tested and analyzed, and the effect of cold storage at 4 ℃on its activity was studied. The results showed that: during the cold storage of anaerobic fermentation inoculum at 4 ℃, the quantity and varieties of methanogens were declined, and their daily anaerobic methanogenesis rate at earlier 15-d stage was obviously lower than that in the untreated control group. However, the cold storage could reduce ammonia nitrogen concentration and contribute to the recovery of anaerobic fermentation activity of inoculure.
