双酶复配同步提取鸭骨素工艺条件 被引量:2

摘要 实验以可溶性固形物含量为测定指标,在鸭骨素提取过程中,研究了碱性蛋白酶、中性蛋白酶和胰蛋白酶复配后同步酶解鸭骨的工艺,优化了复合酶的最佳配比和酶解工艺参数。通过采取双酶(碱性蛋白酶-中性蛋白酶)同步提取的方法,和通过正交试验,优化了双酶添加比例、加酶量、酶解温度、酶解时间和液料比对鸭骨素得率的影响。结果表明,当双酶比例(碱性蛋白酶∶中性蛋白酶)为4∶1,加酶量为2%,酶解温度为60℃,酶解时间为4h,液料比为60g/100g时,在此最佳工艺条件下制得的酶解液中总可溶性固形物含量为32.2%,提取效果最佳。 Use soluble solids content as index,alkaline protease, neutral protease and trypsin complex synchronous enzymatic hydrolysis of duck bone is studied. Optimize the best ratio of compound enzymes and enzyme hydrolysis process parameters. By taking double enzymes (alkaline protease, neutral protease) synchronous extraction method, through orthogonal experiment, optimize the double enzyme addition ratio, the effects of adding amount of enzyme, the enzymatic temperature, enzymatic time and liquid to solid ratio on duck bone yield. The results show that when double enzymes ratio is 4 : 1, enzyme amount is 2%,enzymatic hydrolysis temperature is 60 ℃, enzymatic hydrolysis time is 4 h, liquid and material ratio is 60 g/100 g, the total soluble solids content of the enzymatic hydrolysate is 32.2 % with the best extraction effect.
