寒、热证候大鼠大肠癌启动时ATP酶和琥珀酸脱氢酶活性的变化 被引量:9

摘要 目的:寒、热证候大鼠大肠癌启动时ATP酶和SDH酶活性的变化。方法:将156只大鼠随机分为寒模组、热模组和空白组。寒模组和热模组分别制作寒证、热证模型。第6周开始于大鼠颈背部注射DMH制作大肠癌模型,同时从寒模组中随机取一半大鼠额外给与反左金丸汤剂灌胃,作为寒模反左金丸组;从热模组中随机取一半大鼠额外给与左金丸汤剂灌胃,作为热模左金丸组。期间定期分批处死大鼠,取肠组织进行HE染色,检测肠组织ATP酶活性、SDH酶活性。结果:大鼠在注射DMH 12周及持续每日冷、热刺激后,寒模组与热模组的SDH和ATP酶活性皆出现先升高再降低的过程,而寒模反左金丸组和热模左金丸组相对平稳,维持在较低水平。结论:寒证模型和热证模型大鼠在大肠癌启动时皆会出现阶段性热证,考虑与肿瘤启动时的炎症发热有关,而对应给与的左金丸和反左金丸对阶段性热证有抑制作用。 Objective :To discuss the changes of the activities of ATP and SDH in rats with cold and heat syndrome. Methods:A total of 156 rats were randomly divided into cold model group, heat model group and blank group. Cold and heat syndrome models were made respectively in the cold and heat models. Starting at the sixth week, the colorectal cancer model was induced by DMH neck injection. At the same time, half of rats were randomly selected from the cold model group and given decoction of Fan Zuojin Pill by garage, as the cold model of Fan Zuojin Pill group. While half of rats were randomly selected from the heat model group and given decoction of Zuojin Pill by garage, as the heat model Zuojin Pill group. During this period, the rats were sacrificed regularly and the large intestine tissues were taken out for HE staining and the activities of ATP and SDH were tested. Results :After DMH injection of 12 weeks and continued daily cold or heat stimulation ,the activities of SDH and ATP of cold model group and heat model group both appeared to increase and then reduced while cold model of Fan Zuojin Pill group and heat model Zuojin Pill group are relatively stable, maintaining in a low level. Conclusion:Rats in cold model group and heat model group appear periodic heat syndrome in colorectal cancer initiation. It is considered to be related to fever associated with inflammation of tumor initiation. And Zuojin Pill and Fan Zuojin Pill could bring inhibitory action to the periodic heat syndrome.
