摘要 目的:建立基于matK基因的高良姜及其同属混伪品的分子鉴别方法。方法:采用通用引物对高良姜样品matK基因进行PCR扩增和双向测序,并从Gen Bank下载10种同属混伪品的matK基因序列。采用Clustal X、MEGA软件进行序列比对分析、计算遗传距离以及构建聚类树。结果:获得的高良姜及其同属混伪品matK基因序列长度为732 bp,变异位点41个。高良姜种内遗传距离为0.000,与混伪品之间的最小遗传距离为0.003。基于matK序列构建的系统发生树显示高良姜样品聚在一起,能直观地与混伪品区分。结论:matK基因可以有效地鉴别高良姜及其同属混伪品。 Objective:To establish a molecular identification method for Alpinia officinarum and its adulterants, Methods:The matK gene of A. officinarum was amplified and sequenced using a pairs of universal primers. The matK sequences of 10 adulterants were downloaded from GenBank. Sequence alignments,genetic distance calculation and philogenetic tree construction were conducted using ClustalX and MEGA software. Results :The length of matK gene from A. officinarum and its adulterants was 732 bp,which included 41 variation sites. The intraspecific distance in A. officinarum was 0.000 and the minimal insterpecific distance between A. officinarum and its adulterants was 0. 003. A. officinarum can be obliviously differentiated from its adulterants in the phylogenetic tree. Conclusion:The matK gene can effectively distinguish A. officinarum and its adulterants.
