白术七物颗粒剂对气阴两虚型慢传输型便秘大鼠结肠组织NO的影响 被引量:9

摘要 目的研究在白术七物颗粒剂干预作用下气阴两虚型慢传输型便秘(STC)模型大鼠结肠组织NO改变情况,发现白术七物颗粒剂在治疗气阴两虚型STC中的可能作用机制。方法将60只大鼠随机分成空白对照组10只、模型组50只。造模大鼠按照2.5 mg/kg剂量皮下注射盐酸吗啡注射液,空白对照组注射等量生理盐水,每天1次,连续注射45 d。皮下注射盐酸吗啡注射液15 d后,灌服(青皮、枳壳、附子,剂量13 g/kg),连续30 d,建立气阴两虚型STC大鼠模型。除10只验证模型建立成功外,将其余40只大鼠分为模型组、白术七物颗粒高、低剂量组、莫沙必利组,每组10只。除空白对照组、模型组外,余治疗组给予相应的药物灌胃。应用ELISA、免疫组化方法检测白术七物颗粒剂对气阴两虚型STC模型大鼠结肠组织NO的含量影响。结果白术七物颗粒剂治疗组大鼠结肠组织中NO阳性表达细胞减少,分布相对较稀疏,平均光密度值降低。结论白术七物颗粒剂治疗气阴两虚型STC的作用机制是通过减少结肠组织中NO的含量来实现的。 Objective To investigate the changes in NO in colon tissues of STC model rats with both Qi and Yin defi- ciency under the intervention of atractylodes seven things granules, and to explore the possible mechanism of action of atrac- tylodes seven things granulesin treating STC with both Qi and Yin deficiency. Methods 60 rats were randomized into the nor- mal group including 10 animals and the model group including 50 animals. Rats in the model group were given morphine hydro- chloride injection at 2.5mg,/kg by subcutaneous injection, and rats in the normal group were injected with equal volume of nor- mal saline,once daily for 45 consecutive days. 15 days after subcutaneous injection of morphine hydrochloride injection, rats were given citrus tangerine, fructus aurantii and aconitum carmichaeli debx. ig at 13g/kg for 30 consecutive days to establish the STC rat model with both Qi and Yin deficiency. After the model was established, the remaining 40 rats in the model group were randomized by gender and weight into the model cohort, atractylodes seven things granules high dose cohort, atractylodes seven things granules low dose cohort, and mosapride cohort each including 10 animals. Animals in the treatment cohorts other than the normal group and model cohort were given ig treatment with the corresponding drug. The effect of atractylodes seven things granules on NO level in colon tissues of STC model rats with both Qi and Yin deficiency was detected by using ELISA and IHC. Results In the atractylodes seven things granules treatment cohorts, the number of cells with active NO expression decreased in the colon tissues, with relatively sparse distribution, and the mean OD value decreased. Conclusion Atractylodes seven things granules acts by decreasing NO level in colon tissues to treat STC with both Qi and Yin deficiency.
