摘要
为了对 1 1 6个 1 0 -mer随机引物的RAPD分子标记进行深入了解 ,探讨分子标记与园艺性状的关系 ,筛选出多态性明显、重复性好、亮度大、分子量也较大 ( 1 0 31bp以上 )的 1 0 -mer随机引物 ,在其 3’端添加一任意碱基 ,对 1 4个红木材料和 2个来源不同的生态型木材料进行 3’ -ERPAD标记分析。发现在S2和S35的延伸引物中能够得到最佳扩增结果的引物对是S2 -G +S2-C和S35-A +S35-C ,它们的扩增结果都含有与各自基本引物相同的差异扩增带 ,这些差异带分别是S2 15 0 0 、S2 10 31和S3530 0 0 、S3590 0 、S355 6 0 。通过与园艺性状比较 ,发现S2 15 0 0
To discuss the relation between molecular markers and horticulture character, primers were screened from the early RAPD analysis with 116 random primers (10 mer) which produce strong repetitive polymorphic bands and whose molecular weight is over 1031bp. Then, a random oligonucleotide was added at the 3'-end of the primers to form ERPAD primers. 3'-ERPAD markers were used to analyze 14 Loropetalum chinense var. rubrum and 2 different Loropetalum chinense ecotypes. The result shows that primer pairs S2-G+S2-C and S35-A+S35-C extended from S2 and S35 could produce the best result. Their amplified patterns contain the same polymorphic bands as the basic random primers. These bands are S2 1500 , S2 1031 and S35 3000 , S35 900 , S35 560 . The correlation analysis shows that S2 1500 maybe correlate to the genes that control leaf color.
