摘要
目的研究乌司他丁对脂多糖(lipopoly saccharide,LPS)刺激后的小鼠肺泡巨噬细胞(AM)表面TREM蛋白表达的影响,并探讨乌司他丁在AM介导的炎症反应中的作用及机制。方法培养肺泡巨噬细胞,分为对照组、LPS干预组、LPS+乌司他丁(LU)组。对照组不予处理,LPS干预组给予100ng/mL LPS,LU组给予100ng/mL LPS+10 000U/mL乌司他丁,之后分别在3h、12h、24h用流式细胞仪(FACS)检测AM表面TREM1、TREM2蛋白的表达水平。ELISA检测AM培养上清液中TNF-α表达水平。结果对照组TREM1、TREM2各时间点的表达差异无统计学意义(P>0.05)。LPS干预组TREM1的表达在3h上升到最高,24h时接近正常;TREM2在3h下降到最低,之后上升。LU组TREM1的表达均明显下降,24h下降至最低,与LPS组对比差异有统计学意义(P<0.05);TREM2的表达均明显升高,在24h升至最高,与LPS组对比差异有统计学意义(P<0.05)。TNF-α的表达对照组各时间点差异无统计学意义(P>0.05);LPS组在3h达到最高,之后下降,24h时接近正常;LU组均明显下降,与LPS组对比差异有统计学意义(P<0.05)。结论乌司他丁可能通过调节TREM蛋白在LPS致AM介导的炎症反应中起重要作用。
ObjectiveTo investigate the effect of ulinastatin for expression of TREM after LPS stimulates the alveolar macrophages(AM)and explore mechanism in AM-mediated inflammatory response.Methods Cultured the alveolar macrophages,which were divided into control group,LPS group,and LU group.LPS group was added LPS at the final concentration of 100μg/mL,and LU group was added LPS at the final concentration of 100μg/mL and ulinastatin the final concentration of 10 000U/mL.The expression level of TREM1 and TREM2at 3h,12 h,24h time points was detected by flow cytometry(FACS).And the concentration of TNF-αwas detected by ELISA method.Results The expression of TREM1 and TREM2at Control group there were no statistical significantly differences in other time points(P〉0.05).The expression of TREM1,TREM2 and TNF-αthat there were statistical significantly differences between LU and LPS group(P〈0.05).The expression of TNF-αat Control group there were no statistical significantly differences in other time points(P〉0.05).Conclusion Ulinastatin can adjust the TREM protein in LPS to AM-mediated inflammatory response plays an important role.
