连黄降浊颗粒对自发性高血压大鼠肾动脉重构的影响 被引量:2

Effect of Lianhuang Turbid Clearance Granules on Renal Artery Remodeling in Spontanceously Hypertensive Rats

摘要 目的:观察连黄降浊颗粒对自发性高血压大鼠(SHR)肾动脉重构的影响,并探讨其作用机理。方法:40只SHR随机分为模型组、益肾化湿颗粒对照组(中药对照组)、盐酸贝那普利对照组(西药对照组)、连黄降浊颗粒低剂量组(低剂量组)、连黄降浊颗粒高剂量组(高剂量组),8只WKY大鼠作为正常血压对照组(WKY组),给予相应浓度和剂量药物。给药8周后,测定大鼠血压、尿微量白蛋白(m Alb)、尿微量白蛋白/尿肌酐(Um Alb/Ucr)、血浆血管内皮生长因子(VEGF)和血管紧张素Ⅱ(AngⅡ);计算肾小管损伤指数(TII)和肾小球硬化指数(GSI),测量肾动脉中膜厚度(MT)、管腔内径(LD),检测VEGF表达,观察其中膜超微结构。结果:8周后,模型组SHR血压、m Alb、Um Alb/Ucr、血浆VEGF和AngⅡ显著升高(P<0.01),肾组织病变严重,肾动脉重构明显。低剂量组和高剂量组SHR血压、m Alb、Um Alb/Ucr、血浆VEGF、AngⅡ、TII和GSI显著下降(P<0.01);肾动脉MT、MT/LD下降和VEGF表达减弱(P<0.01);肾动脉血管平滑肌细胞(VSMC)增生、肥厚,内皮细胞脱落,中膜断裂改善,且高剂量组优于低剂量组。西药对照组SHR血压和m Alb、Um Alb/Ucr显著降低(P<0.01);TII、GSI,肾动脉VEGF表达和MT/LD有所下降(P<0.05),效果均不如高剂量组。结论:连黄降浊颗粒能够降低SHR血压、减少尿蛋白、保护肾功能、改善肾组织病变,并抑制SHR肾动脉VEGF过度表达、AngⅡ分泌,逆转其肾动脉重构。 Objective :To study the effect of Lianhuang Turbid Clearance Granules (LHTCG) on Renal Artery Remodeling in SHR,and explore its mechanism. Methods:40 SHRs were divided randomly into five groups: model group, Beneficial kidney and rerolving dampness granules control group ( Chinese medicine group) , Benazepril Hydrochloride control group ( Western medicine group), low -dosage LHTCG group (low -dosage group) and high -dosage LHTCG group (high -dosage group), 8 WKY rats were used as normotensive control group ( WKY group). Medicine was given to each group according to the designed concentration and dosage. Blood pressure, micromalbuin (mAlb) , urine micromalbuin/urine creatinine (UmAlb/Ucr) , plasma vascular endothelial growth factor (VEGF) and angiotensin II (Ang II ) of all rats were measured at 8th week; Tubulointerstitial injury index (TII) and glomeru- losclerosis index (GSI) were calculated;Mesolamella thiekness( MT), lumen inner diameter (LD) and VEGF expression of renal arteries were detected. Ultrastrnctural changes of renal artery were observed. Results:After 8 weeks, blood pressure, mAlb, UmAlb/ Ucr, plasma VEGF and Ang II of model group were increased significantly ( P 〈 0.01 ) , kidney tissue disease and renal artery remod- eling obvious; Blood pressure, mAlb, UmAlb/Uer, plasma VEGF, Angll , TII, GSI of low-dosage group and high-dosage group were decreased significantly (P 〈 0. 01 ) ; MT, MT/LD and VEGF expression of renal artery were decreased (P 〈 0.01 ) ; Vaseularsmooth muscle ceils (VSMC) proliferation, hypertrophy, endothelial cell loss and membrane rupture of the renal artery were improved, and high -dosage group better than low -dosage group. Blood pressure, mAlb,UmAlb/Ucr of Western medicine group were reduced significantly (P 〈 0.01 ) ; TII, GSI, renal artery VEGF expression and MT/LD of renal artery were decreased (P 〈 0.05 ), but the effect is not as good as even LHTCG. Conclusion: Blood pressure and proteinuria of SHR can be reduced, renal function can be protected, pathological changes of renal tissue can be improved, SHRs' renal artery VEGF overexpression and AngII secretion were inhibited by LHTCG, and renal artery remodeling was reversed.
