miR-370在肝癌组织中的表达及与临床病理和预后的关系

Expression of miR-370 and its relation between clinical pathology and prognosis

目的本研究探讨肝癌组织中miR-370的表达,分析其与临床病理和预后的关系。方法收集90对肝癌组织和相应的癌旁组织标本,20例正常肝组织标本为对照,分析不同组织中miR-370的表达水平与临床病理指标之间的关系,并对其中40例随访病例进行了生存分析。结果 Real-time PCR结果表明与正常肝组织相比,miR-370在癌旁组织中表达显著下降,在肝癌组织的表达进一步下降。对90对标本对应病例的临床资料的分析,结果显示miR-370的表达与肿瘤大小、TMN分期、是否血管侵犯、有无肿瘤卫星灶、是否包膜侵犯等肝癌恶性指标密切相关。Kaplan-Meier生存分析表明,低miR-370表达的肝癌患者总体生存时间短于高表达者,结果有统计学意义。结论 miR-370在肝癌组织中的表达与正常肝组织相比显著下调,miR-370低表达水平与肿瘤恶性程度和预后密切相关。

Objective To investigate the expression of the miR- 370 in hepatocellular carcinoma,and analyze the its relation between clinical pathology and prognosis. Methods We collected 20 normal human liver tissues,90 liver cancer tissues and corresponding para-carcinoma tissues,expression of miR-370 in these tissues and its relation between clinical pathology and prognosis were studied.Results Real-time PCR showed that miR-370 expression was substantially repressed in the surrounding non-tumorous livers from HCC patients compared with healthy human livers,and was further reduced in HCC tissues. miR-370 expression was significantly correlated with aggressive pathologic characteristics including larger tumor size,advanced tumor stages,presence of venous invasion,tumor microsatellite formation and presence of capsular invasion. Kaplan- Meier analysis revealed that lower miR-370 levels were correlated with shorter overall survival in HCC patients. Conclusion The miR-370 was consistently down-regulated in HCCs relative to normal liver tissue. Down- regulation of miR- 370 is associated with aggressive tumor behavior and short survival in HCC patients.
