高效液相色谱检测运动饮料中的人工合成色素 被引量:7

Analysis of Synthetic Colorants in Sports Drink by High Performance Liquid Chromatography

摘要 建立一种同时测定运动饮料中柠檬黄、苋菜红、胭脂红、日落黄、诱惑红、亮蓝等6种人工合成色素的固相萃取-高效液相色谱(SPE-HPLC)方法。样品前处理采用固相萃取小柱进行富集和净化,淋洗液选择甲酸-水溶液,洗脱液选择氨化甲醇,氮气吹干后用水复溶上机检测。采用Waters RP C_(18)色谱柱(4.6 mm×250 mm,5μm)分离,以甲醇-乙酸铵水溶液(20.0 mmol/L)作为流动相,用PDA检测器检测,外标法峰面积定量。结果表明,6种人工合成色素在2.00μg/m L^32.00μg/m L浓度范围内线性良好,相关系数r2为0.99,检出限在0.04μg/m L^0.15μg/m L,定量限为0.12μg/m L^0.50μg/m L,加标回收率达到85.6%~92.4%。该方法具有前处理简单、净化效果好、灵敏度高和检测速度快的优点,适用于运动饮料中人工合成色素的分离和定量检测。 A rapid and effective method was established for the determination of six synthetic colorants,including tartrazine,amaranth,carmine,sunset yellow,allura red,and brilliant blue in sports drink by solid-phase extraction coupled with reversed-phase high performance liquid chromatography(SPE-HPLC). The samples were concentrated and purified by solid phase extraction column,and the leaching solution of formic acid aqueous solution,the eluent of amonium methanol The elution liquid was dried under nitrogen,and detected after re-dissolution with water. The separation of targeted compound was performed on a Waters RP C18 chromatographic column(4.6 mm×250 mm,5 μm)using ammonium acetate aqueous solution(20.0 mmol/L)as mobile phase,with PDA detector and external standard method peak area quantification. The linear range of 6 synthetic colorants was in the range of 2.00 μg/m L-32.00 μg/m L with a correlation coefficient of 0.99. The detection limit was 0.04 μg/m L-0.15 μg/m L and the quantitative limit was 0.12 μg/m L-0.50 μg/m L. The recovery rate was 85.6 %-92.4 %. The method has the advantages of simple pretreatment,good purifying effect,high sensitivity and fast detection rate,and is suitable for the separation and quantitative detection of synthetic colorants in sports drink.
