一种硫酸酯化灰树花多糖的免疫活性研究 被引量:6

摘要 灰树花营养价值丰富。灰树花多糖作为一种生物大分子活性物质,表现出良好的抗病毒、预防肿瘤及免疫调节等功能。灰树花子实体中水不溶多糖占据了灰树花多糖的较大比例,硫酸酯化可使其具有良好的水溶性,为实现灰树花多糖资源的充分利用,运用三氧化硫三甲胺盐法制得水不溶性硫酸酯化多糖(S-GFP),其得率约为156%;红外光谱显示,硫酸基主要取代多糖S-GFP的C6位,并有部分双取代存在;测得S-GFP的硫酸基取代度为1.42,总糖平均含量为60.04%;凝胶渗透高效液相色谱显示S-GFP为分子量155 k Da的均一组分。S-GFP免疫活性研究表明,在一定浓度和时间范围内,S-GFP能显著提高RAW264.7细胞的增殖能力,并呈剂量依赖,具有显著的免疫增强活性。 Grifola frondosa is rich in nutritional value. Polysaccharide active substance as a biological macromolecules,has showed good anti-virus,prevent cancer and immunomodulatory functions. Grifola frondosa fruiting bodies of water-insoluble polysaccharide occupy a larger proportion of polysaccharide,sulfated polysaccharide could improve its biological activity,to achieve the full advantage of polysaccharide resources. The sulfur trioxide trimethylamine salt-DMF method was taken to sulfate GFP to S-GFP,a water-soluble sulfated polysaccharide prepared S-GFP,its yield wasabout 156 %,the average total sugar content of 60.04 %,a degree of substitution of sulfate groups 1.42,and mainly for the C6-substituted,di-substituted and some exist,a molecular weight of 155 k Da. The results showed that in a certain range of concentrations and time,S-GFP could significantly improve the proliferation and phagocytic activity of RAW 264.7 cells. S-GFP has a high immunomodulatory activity.
