多指标综合评分优选小儿清肺止咳巴布膏醇提工艺

Optimization of ethanol extraction technology for Xiaoer Qingfei Zhike cataplasm by multi-index comprehensive evaluation method

摘要 目的:通过多指标综合评分法优选小儿清肺止咳巴布膏醇提工艺,确定最佳工艺参数。方法:以处方工艺中麻黄碱、隐丹参酮的提取率及浸出物含量为指标,通过多指标综合评分法,考察醇浓度、溶剂倍量、提取时间及提取次数对小儿清肺止咳巴布膏醇提工艺的影响。采用HPLC法测定麻黄碱和隐丹参酮的含量,流动相分别为乙腈-0.1%磷酸水溶液(5∶95)和甲醇-水(77∶23),检测波长分别为210和260 nm。结果:通过正交实验优选的最佳提取工艺:加15倍量70%乙醇提取2次,第1次加8倍量溶剂提取2 h,第2次加7倍量溶剂提取1 h。麻黄碱和隐丹参酮的提取率及浸出物得率分别为74.44%,85.23%和39.57%。结论:采用多指标综合评分法优选的的醇提工艺科学合理,有效成分提取率高。 Objective:To optimize the ethanol extraction technology for Xiaoer Qingfei Zhike cataplasm,and establish the optimum technological parameters by multi-index comprehensive evaluation method. Methods:With the yield of extraction and extraction rates of ephedrine and cryptotanshinone as indexes,using multi-index comprehensive evaluation method,the effects of the concentration and amount of ethanol,extraction time and extraction times on the ethanol extraction process were investigated. The contents of ephedrine and cryptotanshinone were determined by HPLC,the mobile phases were composed of acetonitrile-0. 1% phosphoric acid solution(5 ∶95) and methanol-water(77∶ 23),and the detection wavelengths were 210 and 260 nm,respectively. Results:The optimal extraction technology was as follows:extraction with 8 times volume of 70% ethanol for 2 h for the first time,and 7 times volume of 70% ethanol for 1 h at last. The extraction rates of ephedrine and cryptotanshinone and the yield of extraction were 74. 44%,85. 23% and 39. 57%,respectively. Conclusion:The optimized ethanol extraction technology for Xiaoer Qingfei Zhike cataplasm by multi-index comprehensive evaluation method is scientific and reasonable with high extraction rates of active components.
