摘要
目的:对紫外分光光度法测定吸收度计算溶血率时波长的选择进行探讨研究。方法:参考《中国药典》2010年版一部附录ⅩⅧB中药注射剂安全性检查法应用指导原则和中药、天然药物刺激性和溶血性研究的技术指导原则进行溶血试验的研究。结果:兔血红细胞破裂后在541nm和576nm处有最大吸收;同一供试品分别采用541mm和576nm测定溶血率,结果无显著性差异。结论:当样品本身在541nm或576nm处有吸收,干扰溶血率计算时可选择另一不干扰波长测定溶血率。
Objective To study the selection of wavelength when UV is used on the hemolytic rate. Methods According to the Guiding Principles of safety tests on traditional Chinese medicine injection in Annex XVIII B, Chinese Pharmacopoeia, 2010 Edition 1, and Technical Guidelines of studies on the irritability and hemolytic activity of traditional Chinese medicine and natural medicine. Re- sults When rabbit red blood cells rupture, there is maximum absorption on 541 nm and 576 nm. There is no significant difference when a same sample was determined on 541 nm or 576 nm and calculate hemolytic rate. Conclusion When a sample have absoption on 541 nm or 576 nm and it interfere in hemolytic rate, we can choose another wavelength to determine the hemolytic rate.
