黑木耳降血脂酸性多糖提取及初步纯化的研究 被引量:3

摘要 为优化黑木耳降血脂酸性多糖的提取工艺,应用单因素试验和响应面分析法进行研究。选择料液比、提取温度、提取时间为主要影响因素,黑木耳多糖的得率(Y_1)和与牛磺胆酸钠(STC)结合率(Y_2)为响应值,进行多个指标同步优化。获得最佳提取工艺参数为:pH为9.0、料液比为1∶80(g/m L)、提取温度为71.5℃、提取时间为2.0 h。在此最佳条件下,多糖得率为3.25%,STC结合率为19.95%。采用弱碱性的大孔阴离子交换树脂D315对该多糖进行初步纯化,有效地对多糖进行脱色和脱蛋白处理,提高多糖纯度,减少多糖活性成分降解,为黑木耳酸性多糖的进一步开发利用提供理论参考。 To study the optimal extraction technology of auricularia acidic polysaccharide with reducing blood lipid, the single factor experiment was utilized combined with response surface methodology.The experimental datas were fitted to create two mathematical models describing yield (Y1) and in vitro binding of STC (Y2)obtained at different levels of critical factors, such as extraction temperature and time and material/liquid ratio. The optimum extraction conditions were as follows: pH 9.0,material to water ratio 1 : 80 (g/mL), extraction tem- perature 71.5 ℃, extraction time 2,0 h.Under these conditions, the yield of polysaccharide was 3.25 %, the binding of STC was 19.95 %.Weak base macroporous anion-exchange resin D315 was used for preliminary purification.The treatment had the effect on discoloration and deproteinized, improveing polysaccharide purity, reducing the degradation of the active ingredient.These can provide a theoretical basis for the further development and utilization of auricularia acidic polysaccharide.
