摘要
本实验室于2012年在河北地区某狐狸场的疑似感染伪狂犬病死亡狐狸中分离到一株伪狂犬病毒(PRV),命名为HB1株,并对其进行了PCR鉴定和特异性血清的间接免疫荧光鉴定。扩增及测序得到的PRV的gC和gE基因序列。与欧美参考株相比,该分离株gE基因编码的氨基酸序列共存在19个替换和2个插入;其gC基因编码的氨基酸序列共存在27个替换,及在第63~69位存在一个GAAASTPA的连续8个氨基酸插入,这与国内近5年来猪群流行的变异株一致。本研究表明PRV变异株也在狐狸群内流行。
A pseudorabies virus (PRV), named HB1, was isolated from fox suspected of being infected with PRV in a fox farm in Hebei region in 2012 by our laboratory, and identified by PCR assay and indirect immunofluorescence assay with specific serum against PRV. Then the gC gene and gE gene were amplified and sequenced for further genetic evolution analysis. Compared with European and American reference strains, the gE gene encoded amino acid sequence of the isolate had 19 substitutions and 2 inserted; the gC gene encoded amino acid sequence of the isolate had 27 substitutions, and a continuous 8 amino acid insertion of GAAASTPA in the position of aa63 to aa69, which were consistent with the prevalence of mutant PRVs isolated from pigs in recently five years in China. In conclusion, these data demonstrated that the variant PRV also prevailed in fox.
