参叶缓痴胶囊对认知功能障碍模型大鼠脑细胞保护作用机制研究

On improving cognitive dysfunction of Shenye Huanchi capsule in delaying brain cell of dementia model rats

目的观察参叶缓痴胶囊对认知功能障碍模型动物空间辨别力的影响及对大脑海马组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的变化,探讨参叶缓痴胶囊对认知功能障碍模型大鼠的脑细胞的保护作用机制。方法采用D-半乳糖复合认知功能障碍模型。采用Morris水迷宫法对模型动物的学习记忆行为进行测试,以动物逃避潜伏期和空间探索行为为观测指标。酶联免疫法观察脑组织SOD、MDA、GSH-PX、IL-6、TNF-α的变化情况。结果参叶缓痴胶囊可以缩短模型动物逃避潜伏时间并提高其空间探索能力;提高脑组织中SOD、GSH-PX的活性,降低MDA、TNF-α与IL-6的含量。结论参叶缓痴胶囊可以改善慢性脑缺血认知功能障碍模型的认知能力,其机制可能与参叶缓痴胶囊对脑细胞的抗氧化作用及抑制脑组织炎性介质有关。

Objective To study the spatial resolution effect of Shenye Huanchi capsule on the model animalcognitive dysfunction and the mechanism of action of Shenye Huanchi capsule in cerebrum hippocampus on SOD, MDA, GSH-PX, IL-6 and TNF-α. To explore the protection mechanismof Shenye Huanchi capsule in delaying brain cellof dementia model rats on improving cognitive dysfunction. Methods In this study, we used the D-galactose complex cognitive dysfunction model.Morris water maze was used to test the learning and memory behavior of rats by measuring the escape latency and spatial probe behavior. The changes of SOD, MDA, GSH-PX, IL-6, TNF-α were observed in brains with enzyme-linked immunosorbent assay（ELISA）. Results Shenye Huanchi capsule can shorten the model animals to escape latency and improve their ability to explore space. Shenye Huanchi capsule can improve the SOD and GSH-PX activity and decrease the MAD, TNF-α and IL-6 concentration. Conclusion Shenye Huanchi capsule can improve the cognitive ability of the model of chronic cerebral ischemia. The mechanism may be related to the anti oxidation of brain cells and the inhibition of brain tissue inflammatory mediators.
