甲巯咪唑致亚临床甲减大鼠模型评价 被引量:5

摘要 目的探讨应用甲巯咪唑(MMI)建立亚临床甲减(SCH)大鼠模型的有效给药剂量及时间点。方法将45只成年雄性Wistar大鼠随机分为3组:正常对照组;MMI低剂量组予MMI 5 mg/(kg·d)灌胃;MMI高剂量组予MMI 10 mg/(kg·d)灌胃,分别于给药后第45、90、120天采血测定甲状腺功能。结果 45 d时MMI高剂量组血清TSH水平与正常对照组比较差异有统计学意义(P<0.05),符合SCH标准的有7只,成功率为63%(7/11)。90 d时MMI低剂量组和MMI高剂量组分别与正常对照组比较:血清TSH水平差异均有统计学意义(P<0.05),90 d时MMI高剂量组符合SCH诊断标准有8只,成功率为66%(8/12),MMI低剂量组符合SCH的诊断标准有6只,成功率为60%(6/10)。120 d时MMI低剂量组和MMI高剂量组分别与正常对照组比较:血清TSH及T4水平差异均有统计学意义(P<0.01)。120 d时MMI高剂量组符合SCH诊断标准有1只,成功率为8%(1/12),MMI低剂量组符合SCH的诊断标准有4只,成功率为33%(4/12)。结论应用MMI可以成功建立SCH大鼠模型。 Objective To investigate the effective dose and time point of methimazole(MMI) on subclinical hypothyroidism(SCH) rat model. Methods Forty five adult male Wistar rats were randomly divided into three groups:normal control group; MMI low dose group received MMI 5mg/(kg·d)gavage; MMI high dose group received MMI 10mg/(kg·d)gavage, respectively to the drug after the 45 days, 90 days and 120 days blood determination of thyroid function. Results 45 days MMI high-dose group, the serum TSH levels compared with normal control group, the difference has statistical significance(P 0.05), accord with the standard of SCH 7 only, the success rate was 63%(7/11). 90 days MMI low dose group and MMI high dose group compared with the normal control group:serum TSH levels were statistically significant difference(P 0.05).At 90 days MMI high dose group in accordance with the SCH diagnostic criteria has 8, the success rate of 66%(8/12), MMI low dose group in accordance with the diagnostic criteria of SCH has 6, the success rate of 60%(6/10). 120 days MMI low and high dose group compared with the normal control group:serum TSH levels and serum T4 were statistically significant difference(P〈0.05). At 120 days MMI high dose group in accordance with the SCH diagnostic criteria has 1, the success rate of 8%(1/12). MMI low dose group in accordance with the diagnostic criteria of SCH has, the success rate of 33%(4/12). Conclusion SCH rat model can be established by using MMI.
