摘要
目的研究Ⅱ型跨膜丝氨酸蛋白酶(typeⅡtransmembrane serine proteases,TTSPs)与非小细胞肺癌(NSCLC)发生发展的关系。方法利用基因表达数据库(gene expression omnibus,GEO)的全外显子基因芯片数据,计算癌组织与癌旁组织的基因相对表达水平,筛选差异表达基因;研究肺癌组织中TTSPs基因的相对表达量,筛查NSCLC中特异性高表达的TTSPs,并分析其在不同分期肺癌中的表达差异;检测肺癌细胞株中TTSPsmRNA的表达。结果TMPRSS4、Malriptase、TMPRSS13在NSCLC中表达显著高于癌旁正常组织(P〈0.001);TMPRSS4的表达在不同分期的肿瘤中差异有统计学意义(P〈0.05);肺癌细胞株中检测到TMPRSs4、Matriptase、TMPRSS13的表达。结论TTSPs中的TMPRSS4、Matriptase和TMPRSS13:(ENSCLC中有特异性高表达,可能与肿瘤的发生发展相关。
Objective To investigate TTSPs expression in non-small cell lung cancer (NSCLC) samples. Methods The expression data of whole exon microarray were downloaded from gene expression omnibus (GEO), and analyzed using R language and packages of bioconductor. The differential expression genes were determined by comparing caner tissues and adjacent normal tissues. TTSPs expression was further analyzed by TNM system. TTSPs mRNA expression in cell lines of lung cancer was analyzed by RT-PCR. Results TMPRSS4, Matriptase and TMPRSS13 expression levels were significantly up-regulated in NSCLC (P 〈 0.0001 vs. normal tissues). TMPRSS4 expression levels seemed to correlate with the severity of disease. And TMPRSS4, Matriptase and TMPRSS13 mRNA expression were detected in the lung cancer cell lines by RT-PCR. Conclusion TMPRSS4, Matriptase and TMPRSS13 were abnormally up-regulated in NSCLC. Our results indicated that TTSPs may contribute to the pathogenesis of NSCLC.
