摘要 椎间盘退变性疾病是临床常见疾病,目前临床的治疗方法只能解除症状,无法从根本上治愈该病.髓核组织工程研究能够重建髓核组织,修复和逆转退变椎间盘.在组织工程研究中,同种异体的髓核细胞常被用作种子细胞,为了使种子细胞在移植后获得更高的活性,更长的存活时间以及更好的功能表达,要对髓核细胞进行一系列的修饰,主要包括:抗凋亡、调节自噬以及永生化修饰.抗凋亡和调节自噬的目的都是使种子细胞在移植后具有较高的活性,能够长期有效地行使功能,从而保证治疗的长期效果.永生化是为了解决髓核细胞传代次数有限,数量不足的问题.本文主要围绕以上三个方面展开综述. Degenerative disc disease is a common clinical disease. The current clinical treatment can only relieve symptoms.However,nucleus pulposus tissue engineering can reconstruct the nucleus pulposus,repair and reverse degenerative disc. In tissue engineering studies,allogeneic nucleus pulposus cells are often used as seed cells. In order to make seed cells obtain higher viability,longer survival time and better functional expression after transplantation,a series of modifications have to be applied to nucleus pulposus cells,including: anti-apoptosis,regulation of autophagy and cellular immortality. The purpose of anti-apoptosis and autophagy regulation is to make seed cells have high viability after transplantation,and to ensure the long-term effect of treatment. Cellular immortality can solve the problem of limited passage number and insufficient cell resources. This article mainly focuses on the above three aspects.
