草苁蓉环烯醚萜苷对大鼠肝纤维化的保护作用及其机制 被引量:2

摘要 [目的]探讨草苁蓉环烯醚萜苷对肝纤维化大鼠肝损伤的保护作用及其机制.[方法]取成年Wistar雄性大鼠随机分为正常对照组、模型对照组、草苁蓉环烯醚萜苷组.皮下注射给予模型对照组和草苁蓉环烯醚萜苷组40%(体积分数)四氯化碳植物油混悬液建立大鼠肝纤维化模型,每周2次,共计5周.草苁蓉环烯醚萜苷组于第6周开始每日连续灌胃给药,剂量为200mg/kg.于第10d采集各组血液及肝组织,利用ELISA法检测血清天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)及总胆红素(TBIL)水平,并利用HE染色、Masson染色、Western blot法观察肝组织形态学变化及α-SMA表达水平.[结果]与正常对照组比较,模型对照组大鼠血清中ALT,AST及TBIL水平均明显增高(P<0.05);HE染色和Masson染色结果显示,模型对照组大鼠肝细胞发生变性、坏死,发生肝纤维化;Western blot检测结果显示,模型对照组大鼠肝组织中α-SMA表达水平明显增高(P<0.05).与模型对照组比较,草苁蓉环烯醚萜苷组ALT,AST及TBIL水平均明显降低(P<0.05),肝纤维化程度明显减轻,肝组织中α-SMA表达水平明显下降(P<0.05).[结论]草苁蓉环烯醚萜苷可通过抑制肝星状细胞的活化来抑制肝纤维化的发生,对肝损伤具有一定的保护作用. OBJECTIVE To study the protective effects and its mechanisms of boschniakia rossica iridoid glycoside on liver fibrosis in rats. METHODS Adult male Wistar rats were randomly divided into the normal control group, model control group and boschniakia rossica iridoid glycoside group. Rat liver fibrosis model was established by subcutaneous injection of suspension of vegetable oil and carbon tetrachloride (40%) 2 times a week for 5 weeks. The rats in Boschniakia rossica iridoid glycoside group were treated by Boschniakia rossica iridoid glycoside (Po, 200 mg/kg per day) continuously since the sixth week, after 10 d-treatment, their blood and liver tissues were harvested. ELISA was used to detect the serum aspartic acid transaminase (AST), alanine aminotransferase (ALT) and total bile acid (TBIL), and HE staining,Masson staining and Western blot were observe liver morphological changes used to and the expression of a-SMA. RESULTS As compared with the normal control group, serum ALT, AST and TBIL levels were significantly increased in the model control group (P〈0.05). HE staining and Masson staining showed that the hepatocytes ofmodel group exhibited degeneration, necrosis and hepatic fibrosis. Western blot showed that the expression of a-SMA was significantly increased in the liver tissue of model control group (P〈0.05). The rats of Boschniakia rossica iridoid glucoside group showed significantly lower levels of serum ALT, AST and TBIL (P〈0.05), markedly reduced liver fibrosis as compared with that in model control group. The expression of α-SMA in Boschniakia rossica iridoid glucoside group was significantly lower than that in model control group (P〈0.05). CONCLUSION Boschniakia rossica iridoid glycoside could inhibit the occurrence of liver fibrosis by inhibiting the activation of hepatic stellate cells, it had certain protective effects to liver damage.
