番茄游离小孢子培养及胚状体诱导研究 被引量:2

摘要 [目的]探讨基因型、花粉发育时期对诱导胚状体的影响和胚状体形成过程中花粉粒的变化特性。[方法]以市场购买的"贼不偷""大黄888""中蔬四号""黄香蕉""顶心红""粉宝莲""宝石"7个番茄品种的花粉粒为材料,以B5为基本培养基,液体培养诱导胚状体。[结果]基因型是影响诱导胚状体的关键因素。供试的7个番茄品种中只有"贼不偷"品种的花粉粒对花粉培养有响应,其余品种无响应;花粉发育时期又是影响诱导胚状体的关键因素。"贼不偷"品种不同发育时期花粉粒对花粉培养的响应有差异,最佳时期是单核中期及单核靠边期;诱导出来的胚状体呈现带有胚柄、多细胞、球形或心形。[结论]试验结果为建立番茄游离小孢子培养体系及其相关机理研究提供了理论依据。 [ Objective] The aim was to study effects of genotype and pollen development period on induced embryoi and the change features of pollen grain in the formation process of embryoid. [ Method ] We took the pollen of "zeibutou" "dahuang-888" "zhongshu-4" "huangxiangjiao" "dingxinhong" "fenbaolian baoshi"varieties as materials, and cultured in B5 liqued medium to induce the embryoid. [ Result ] The genotype was one of the key factor influencing the inducing embryo, Only the pollens of "zeibutou" varieties effectively responsed to the pollen culture. The stage of pollen development was one another key factor influencing the inducing embryoid. The early and late uninuclear stages were the best stage for pollen culture. The induced embryoid displayed suspensur-bearing, muhicellular, global or heart shapes. [ Conclusion ] The results provide theoretical basis for establishing the pollen culture system and related mechanism research.
