Th17细胞及相关细胞因子在老年大肠癌患者中的作用

摘要 目的探讨Th17细胞及相关细胞因子在老年大肠癌患者中的作用。方法选取48例65岁以上的大肠癌患者(A组)、32例65岁以下的大肠癌患者(B组)、同期健康体检的48例65岁以上健康者(C组)、32例65岁以下的健康者(D组),分别用流式细胞仪和Bio-Plex悬浮芯片技术检测外周血中Th17细胞比例及IL-17A、IL-17F、IL-21、IL-22和IL-23水平,用免疫荧光共聚焦显微镜分析大肠癌患者癌组织和癌旁组织中IL-17表达水平。结果大肠癌患者外周血中Th17细胞比例高于健康者(P<0.05),老年患者高于中青年患者(P<0.05)。大肠癌患者外周血中IL-17A、IL-21、IL-22和IL-23水平高于健康者(P<0.05),老年患者IL-17A和IL-23水平高于中青年患者(P<0.05),而老年患者IL-21和IL-22水平与中青年患者比较无明显差异(P>0.05);IL-17F水平在各组人群中无明显差异(P>0.05)。癌组织IL-17表达高于癌旁组织(P<0.05),老年患者IL-17表达水平明显高于中青年患者(P<0.05)。结论 Th17细胞与大肠癌及年龄有相关性,可能在老年大肠癌的发生发展中发挥作用。 Objective To investigate the role of T h l7 cells and related cytokines in elderly patients with colorectal cancer. Methods 48 patients with colorectal cancer over 65 years old were enrolled in group A, 32 patients with colorectal cancer under 65 years old in group B, 48 norm over 65 years old in group C and 32 norm under 65 years old in group D; flow cytometry and Bio-Plex suspension chip technology were applied in detecting the proportion of Thl7 cells and the levels of IL-17A、IL-17F、IL-21、 IL-22 ^ IL-23 in peripheral blood of the patients and the norm while the immunofluorescence confocal microscope was applied in detecting the levels of IL-17 in cancer tissue and adjacent tissue. Results The percentage of Thl7 cells in peripheral blood of the patients with colorectal cancer was higher than that of the norm (P〈 0.05), and that in the elderly patients was higher than that in the young patients (P 〈0.05); the levels of IL-17 A,IL-21, IL-22 and IL-23 in peripheral blood of the patients with colorectal cancer were higher than those of the norm (P〈0.05); the levels of IL-17 A, IL-23 in elderly patients were higher than those in young patients (P〈0.05); there existed no significant difference in the levels of IL-21, IL-22 between the elderly patients and the young patients (P〉0.05), and no obvious difference in the level of IL-17F among the 4 groups; the expression of IL-17 in cancer tissue was higher than that in adjacent tissues (P〈0.05), and that in the elderly patients was higher than that in young patients (P〈0.05). Conclusions Thl7 cells are correlated to colorectal cancer and the age of patients, which may play a certain role in the occurrence and progress of colorectal cancer in elderly patients.
