摘要
目的明确杜仲治疗蛛网膜下腔出血后脑血管痉挛发生的有效性,及其与内皮素-1的关系。方法 72只日本大耳白兔随机分为3大组,正常组仅做枕大池穿刺注生理盐水。SAH组和杜仲组采用枕大池二次注血法制作迟发性脑血管痉挛模型,杜仲组给予杜仲煎剂每日3 ml/kg灌胃,2次/日,造模成功后在相应的时间段分两批处死,半数甲醛灌注取基底动脉,切片HE染色后,观察形态变化,计算周长,半数处死后快速取脑分离出基底动脉用Western blot方法测定ET-1含量。结果 SAH组较正常组有明显基底动脉血管痉挛,而杜仲组较SAH组血管痉挛有明显缓解。正常组中基底动脉仅表达少量ET-1,SAH组中基底动脉ET-1表达较正常组明显升高(P<0.05),杜仲组较SAH组在蛛网膜下腔出血3~7 d时基底动脉中ET-1含量明显降低(P<0.05)。结论杜仲能有效的缓解蛛网膜下腔出血后引起的脑血管痉挛,并且是通过影响基底动脉中ET-1含量来实现的。
Objective To clear the effect of Eucommia ulmoidesthe on delayed cerebral vasospasm(DCVS) following experimental sub-arachnoid hemorrhage occurred and its relationship with endothelin-1. Methods 72 Japanese big ear rabbits were randomly divided into three groups. The normal group received only pillow big pool puncture saline injection. The SAH group of eucommia ulmoides and the pillow secondary injection blood rule1 of law as a large pool of late-onset cerebral vasospasm model, eucommia bark of eucommia ulmoides group was given decoction daily 3 ml/kg to fill the stomach, 2 times/day, after the success of the building in the correspond-ing period was divided into two group of death, half formaldehyde take basilar artery perfusion, slice, after HE dyeing observation form change, calculate the perimeter, after half to death quickly cerebral basilar artery was isolated using Western blot method for deter-mining content of ET-1. Results SAH group was compared with normal way obvious basal artery vasospasm, eucommia ulmoides and the SAH group vasospasm had eased significantly. Basilar artery in the normal way to express only a few ET-1, basal artery ET-1 expression in SAH group increased significantly in the normal group(P〈0.05), eucommia bark from SAH group 3 to 7 days in subarachnoid hemorrhage ET-1 in the basilar artery significantly decreased(P〈0.05). Conclusion Eucommia bark can effectively alleviate cerebral vasospasm after subarachnoid hemorrhage, and has influence on the ET-1 in the basilar artery.
