痛风性关节炎患者中长链非编码RNA的表达谱研究 被引量:11

Study on the expression profile of long non-coding RNAs in gouty arthritis

摘要 目的分析长链非编码RNA(lncRNAs)在痛风性关节炎患者中的表达情况,探讨lncRNAs在痛风性关节炎发病中的分子机制。方法采用lncRNAs表达谱芯片技术,检测痛风性关节炎患者和健康体检者PBMCs中的差异表达lncRNAs和mRNAs,对差异表达的基因进行GO及Pathway分析,并运用实时荧光定量PCR技术对差异表达的lncRNAs进行验证,采用Kruskal-Wallis检验或Mann-Whitney检验进行统计学分析。结果芯片数据结果获得了具有2倍以上差异表达的lncRNAs共1 815条,差异表达的mRNAs有971条。GO分析发现,差异表达的基因主要参与蛋白结合、转录调节、代谢、信号转导等。Pathway分析显示它们主要参与细胞因子和炎症反应途径、糖皮质激素受体调节网络、DREAM信号通路等。检测了4个lncRNAs在痛风性关节炎患者中的表达与基因芯片一致,其中AJ227913是表达差异最为明显的lncRNAs。结论多条lncRNAs在痛风性关节炎患者PBMCs中异常表达,提示它们在痛风性关节炎发生、发展过程中可能发挥着重要作用。 Objective To analyze the expression profile variation of long non-coding RNAs (lncRNAs) in gouty arthritis (GA) patients and explore the role of lncRNAs in the pathogenesis of GA. Methods The peripheral blood mononuclear cells of GA patients and healthy controls (HC) were used to detected for differently expressed lncRNAs by microarray. The role of lncRNAs were predicted with GO and Pathway analysis. Then the results were verified by Real time-polymerase chain reaction (PCR). Kruskal-Wallis test or Mann-Whitney test were used for statistical analysis. Results A total of 1 815 lncRNAs and 971 mRNAs were detected, which was more than 2-fold differentially expressed in GA patients. Bioinformatics analysis found that GO term enrichment included protein binding, regulation of transcription, metabolism, signal transduction, et al. and might involve in cytokines and inflammatory response, glucocorticoid receptor regulatory network, DREAM signaling pathway and so on. The expression of four lncRNAs was estimated by Real time-PCR which was consistent with that of microarrays. Among these, AJ227913 was the most aberrantly expressed lncRNAs. Conclusion Several lncRNAs expression is changed significantly in GA patients in comparison with HC, which implies that those different lncRNAs may have an important role in the development and progression of GA.
