摘要
利用响应面法优化藤本豆豆荚总黄酮提取工艺并进行体外抗氧化活性的评价。通过单因素试验选定提取时间、提取温度、料液比和乙醇浓度为考查因素,利用Box-Behnken进行试验设计,回归和方差分析结果显示,回归模型极显著(p<0.000 1),试验方法可信度较高。藤本豆豆荚总黄酮提取的最佳工艺如下:时间5 h,温度90℃,液料比1∶20(g/mL),乙醇浓度60%。在此条件下藤本豆豆荚总黄酮的得率为6.42%。藤本豆豆荚总黄酮对ABTS+·(IC_(50)=87.86μg/mL)和DPPH自由基(IC_(50)=71.91μg/mL)具有一定的清除作用,对Fe^(3+)具有较好的还原能力(相关系数R^2=0.994),藤本豆豆荚总黄酮表现一定的体外抗氧化活性。
The response surface methodology was utilized to optimize extraction process of total flavonoids from Perennial lablab sp. hull (FPLH) and its antioxidant activity were studied. Through single experiments,extraction time, extraction temperature ,ratio of liquid to solid and ethanol concentration were selected for Box-Behnken central composite designing.The results of regression and ANOVA revealed that the regression model for total flavonoids yield exhibited tremendous significance (p〈0.000 1). The optimal extraction process were time 5 hours , temperature 90℃, ratio of liquid to solid 1∶20(g/mL) and ethanol concentration 60%.The yield of FPLH was up to 6.42 %.The antioxidant tests results indicated that scavenging capacity (IC50)of total flavonoids towards ABTS+&#183;and DPPH·;were 87.86μg/mL and 71.91μg/mL, respectively. FPLH showed strong reducing power towards Fe^3+(R^2=0.994). FPLH showed strong antioxidant activity in vitro.
