摘要
目的探讨金钗石斛多糖(NDP)对新生大鼠大脑皮质胶质细胞-神经元混合培养体系的作用。方法制备新生大鼠大脑皮质胶质细胞-神经元混合培养体系,分别用NDP与脂多糖(LPS)刺激并观察细胞生长情况,采用Western blot检测体系中炎症相关因子IL-1β、TNF-α、COX-2、IL-6的蛋白表达及相关调控信号p-NF-κB、p-p38、p-JNK、p-ERK的水平。结果LPS作用于大鼠胶质细胞-神经元混合培养体系时,炎症相关因子IL-1β、TNF-α、COX-2、IL-6的蛋白表达及相关调控信号pNF-κB、p-p38、p-JNK、p-ERK的水平明显升高。然而,NDP明显抑制了LPS诱导的系列变化。结论 NDP可阻遏LPS诱导的新生大鼠皮质胶质细胞-神经元混合培养体系中小胶质细胞和星形胶质细胞激活,从而保护神经元。
AIM To investigate the effect of noble dendrobium polysaccharides (NDP) on mixed cultured cerebral cortex glial cells and neuron of newborn rats. METHODS The primary cultures of newborn rat cortical neurons and glial cells were established and the existence of the neurons, astrocytes and microglia were verified,respectively. NDP and lipopolysaccharide (LPS) were given to mixed cultures. The protein expression of IL-1β, TNF-α, COX-2, IL-6 and the levels of p-NF-κB, p-p38, p-JNK, p-ERK were detected by Western blot. RESULTS When LPS were given to mixed cultures,the protein expression of IL-1β, TNF-α, COX-2, IL-6 and the levels of p-NF-κB, p-p38, p-JNK, p-ERK were significantly increased. However, NDP inhibited a series of changes induced by LPS in mixed cultured cerebral cortex glial cells and neuron of rats. CONCLUSION NDP protects neuron against LPS-induced activation of microglia and astrocytes in mixed cultured cerebral cortex glial cells and neuron of rats.
