摘要
目的建立根痛合剂中葛根素和芍药苷含量测定的高效液相色谱法(HPLC)。方法采用依利特Elite-C_(18)色谱柱(200mm×4.6 mm,5μm);流动相:乙腈-甲醇-水(15∶15∶70);流速:1.0 m L·min^(-1);检测波长:250 nm,柱温:25℃。结果葛根素在0.2676~2.676μg范围内线性关系良好(r=0.999 9),平均加样回收率为100.1%(n=6),RSD为0.8%,芍药苷在0.2024~2.024μg范围内线性关系良好(r=0.999 8),平均加样回收率为99.2%(n=6),RSD为1.1%。结论该试验所建立的方法简单可行,重复性好,可以用于控制根痛合剂的质量。
OBJECTIVE To establish a HPLC method for the determination of puerarin and paeoniflorin in Gen Tong Mixture.METHODS Elite C18column( 200 mm×4.6 mm,5 μm) was used for the HPLC analysis with mobile phase consisted of acetonitrile,methanol and water( 15 ∶15 ∶70). The flow rate was 1.0 m L·min^-1 with the detecting wave length of 250 nm,and the column temperature was set at 25 ℃. RESULTS The calibration curve of puerarin was linear within the range of 0. 2676- 2. 676 μg( r = 0. 999 9); The average recovery was 100.1%( n = 6) with the RSD of 0.8%. The calibration curve of paeoniflorin was linear within the range of 0.2024- 2.024 μg( r = 0.999 8); The average recovery was 99.2%( n = 6) with the RSD of 1.1%. CONCLUSION This method is simple and reproducible which can be used for the quality control of Gen Tong Mixture.
