丁香叶中鞣质的提取与含量测定

摘要 目的:本文以丁香叶为原料,对丁香叶中鞣质的提取和含量测定,研究结果为综合利用与深入开发提供了一定理论基础。方法:丁香叶中提取鞣质在单因素实验的基础上,采取正交的实验的方法,用提取温度,溶媒倍数,提取时间,丙酮浓度为参考,每个参考有3个水平,最后用分光光度法测定鞣质含量作为综合评价指标。结果:丙酮浓度70%,温度55Ⅵ,时间2h,溶媒倍数14倍为最佳提取条件,含量测定以没食子酸为标准样品建立线性方程,y=92.049x+0.0187,在浓度0.001~0.01mg/m L呈线性关系,含量测定鞣质的含量为0.893%。结论:综上所述,丁香叶中鞣质的提取因素影响为提取温度>提取加热时间>溶媒倍数>丙酮浓度。采取热回流提取方法操作简单,提取效果好。 In this paper, clove leaf as raw material, extraction and content determination of tannins in clove leaf, the results of the study for the comprehensive utilization and in-depth development provides a certain theoretical basis. Clove leaf tannin extracted on the basis of single factor experiment, adopts the method of orthogonal experiment, extraction temperature, solvent ratio, extraction time,concentration of acetone as the reference, each reference has three levels, the spectrophotometric method determination of tannin content was used as the comprehensive evaluation index. 55 ℃, 70% acetone concentration, temperature, time 2 h, solvent ratio 14 times as the best extraction conditions and content determination of gallic acid as standard sample to establish linear equation, y = 92.049 +0.0187 x, the concentration of 0.001 ~ 0.01 mg/m L linear relationship, determine the content of tannin content was 0.893%.
