摘要
目的探讨苗药香囊对小鼠肺组织中核因子-κB(NF-κB)(P65)表达的影响。方法昆明种雄性小鼠80只,随机分为8组,空白对照组(A组)、匹多莫德组(阳性对照组,C组)、苗药香囊组(E组)、匹多莫德+苗药香囊组(G组)、冷刺激组(B组)、匹多莫德冷刺激组(D组)、苗药香囊冷刺激组(F组)、匹多莫德+苗药香囊冷刺激组(H组)。预防用药4周后,B、D、F、H四组予冷刺激[(4±1)℃暴露3 h]4 d制造冷刺激免疫低下模型。采用免疫组织化学染色、实时荧光定量PCR(RT-PCR)和Western blot法检测小鼠肺组织中NF-κB(P65)的表达。结果与A组比较,E、C、G组小鼠肺泡上皮细胞NF-κB(P65)表达增加,差异有统计学意义(P<0.05)。与B组比较,F、D、H组小鼠肺泡上皮细胞NF-κB(P65)表达增加,差异有统计学意义(P<0.05)。结论苗药香囊能够上调小鼠肺组织中NF-κB(P65)的表达,增强呼吸道免疫力。
Objective To study the effect of Miaoyao sachet on NF-κB( P65) expression in lung tissues of mice.Methods Eighty Kunming species male mice were randomly divided into 8 groups,normal control group( Group A),pidotimod group( Group C,positive control),sachet group( Group E),sachet plus pidotimod group( Group G),cold stimulation group( Group B),pidotimod cold stimulation group( Group D),sachet cold stimulation group( Group F),and sachet plus pidotimod cold stimulation group( Group H).Four weeks after prophylaxis,cold water stimulation[( 4 ± 1) ℃ 3 h exposure for 4 days]was performed in Groups B,D,F and H to establish immunodeficiency models.The expression of NF-κB( P65) was assessed using immunohistochemistry,quantitative real-time PCR and Western blot.Results Compared to Group A,NF-κB( P65) were significantly increased in the mice bronchial epithelial cells and alveolar epithelial cells of( P〈0.05).Compared to the Group B,NF-κB( P65) were significantly increased in Group D,F and H( P〈0.05).Conclusion Miaoyao sachet can improve the expression of NF-κB( P65) in lung tissues of mice,strengthen the immune of respiratory tract.
