摘要
目的比较脱水淫羊藿素与金雀异黄酮对受体状态不同的人乳腺癌细胞株增殖的影响。方法运用MTT法研究不同浓度的脱水淫羊藿素与金雀异黄酮对MCF-7、MDAMB231细胞增殖的影响,并以雌激素受体阻断剂ICI182,780和雌激素受体激动剂DPN、PPT为工具药来评价脱水淫羊藿素和金雀异黄酮雌激素样作用与雌激素受体(ERs)的关系。结果脱水淫羊藿素和金雀异黄酮终浓度为10^(-13)~10^(-7)mol/L时,MCF-7细胞株增殖均随浓度升高而增加,呈一定的剂量相关性;而各受试药物任何浓度对MDA-MB231细胞株均无显著增殖效果。加入终浓度为10^(-8)mol/L的ICI182,780时,MCF-7细胞增殖作用被拮抗,MDA-MB231细胞增殖无明显变化。加入终浓度为10^(-8)mol/L的ERα受体通路激动剂PPT时,MCF-7细胞增殖率明显增高(P<0.05),MDA-MB231细胞增殖无明显变化。加入终浓度为10^(-8)mol/L的ERβ受体通路激动剂DPN时,MCF-7和MDA-MB231细胞增殖均无明显变化。结论脱水淫羊藿素与金雀异黄酮在一定剂量范围内均有雌激素样作用,两者作用相当,体外实验促进MCF-7细胞增殖作用可能主要是由ERα介导,而非ERβ介导。
Objective It is to compare the effect of Anhydroicaritin and Genistein on proliferation of human breast cancer cell lines with different receptor situation. Methods With MTT cell proliferation method,we studied the effect of different concentration of Anhydroicaritin and Genistein on the proliferation of MCF-7 and MDA-MB231,and ICI 182. 780( estrogen receptor antagonist),DPN( ERβreceptor pathway agonist) and PPT( ERαreceptor pathway agonist) were used as the tool drugs to evaluate the relationship between estrogenic effect of Anhydroicaritin and Genistein with estrogen receptors. Results When the final concentration of the Anhydroicaritin was 10^-13- 10^-7mol / L,the proliferation of MCF-7 was promoted with the increasing of concentration,and that was dose related,which was as the same as the Genistein. But any concentration of tested drugs had no significant effect of promoting the proliferation of MDA- MB231. When adding the final concentration of 10^-8mol / L ICI182,780,the proliferation of MCF-7 in Anhydroicaritin and Genistein were reduced significantly( P〈0. 05). When adding the final concentration of 10^-8mol / L DPN,the proliferation of MCF-7 in Anhydroicaritin and Genistein was promoted significantly( P〈0. 05). And the proliferation of MDA-MB231 had no change. When adding the final concentration of 10^-8mol / L PPT,the proliferation of MCF-7 in Anhydroicaritin and Genistein had no significant change( P〈0. 05). And the proliferation of MDA-MB231 had no change. Conclusion Anhydroicaritin and Genistein both had estrogenic effect in the certain dose range and their estrogenic effects were similar. The Anhydroicaritin and Genistein's effect of promoting the proliferation of MCF- 7 might be mediated by ERα,but not mediated by ERβ.
