摘要 目的 研究远志总皂苷(Polygala total saponins,PTS)抗皮肤光老化作用。方法 采用体外清除羟自由基、超氧自由基、DPPH氧自由基等实验,考察远志总皂苷对氧自由基的影响;建立小鼠皮肤光老化模型,观察远志总皂苷对小鼠皮肤病理变化的影响。结果 在体外抗氧化实验中,远志总皂苷乳液在6.0、12.0mg/m L浓度具有显著的清除羟自由基(P〈0.05,P〈0.001)、超氧自由基(P〈0.01)和DPPH自由基的作用(P〈0.001)。在光老化实验中,B组(模型+基质茶油)成功建立了小鼠皮肤光老化模型,与B组相比较,D组(照射前30min给药,模型+PTS 6mg/m L剂量组)小鼠皮肤表皮致密增厚,弹力纤维排列均匀致密,胶原纤维显著增生,致密,分布均匀,未见炎性细胞浸润,治疗效果最为显著;G组(照射后30min给药,模型+PTS 12mg/m L剂量组)小鼠皮肤表皮增厚,弹力纤维排列均匀,胶原纤维有一定增生,分布均匀,未见炎性细胞浸润,表明PTS有显著治疗效果。结论 远志总皂苷具有显著抗氧化活性及对抗光老化作用。 Objective To study the anti-aging effect of total saponins of Polygala tenuifolia (PTS). Methods The effects o f total saponins o f Polygala tenuifolia on oxygen free radicals were investigated by the methods of scavenging hydroxyl radical, superoxide radical and DPPH oxygen free radicals in vitro. The photoaging model of mice skin was established to observe the effect of total saponins of Polygala tenuifolia on mouse skin pathology. Results In the experiment of antioxidation in vitro, the total saponins o f Polygala tenuiflora significantly scavenged hydroxyl free radicals (P〈0.05, P〈0.001) and superoxide radical (P〈0.01) at 6.0 and 12.0mg/mL, DPPH &ee radicals (P〈0.001). In the light aging experiment, the light aging model of mice skin was established successfully in group B (model + medium tea oil). Compared with group B,the density of the skin epidermis in D group (30 min before irradiation, 6mg/mL PTS) was thickened, the elastic fibers were arranged uniformly and tightly, the collagen fibers proliferated significantly and densely, and the inflammatory cells infiltration was not found. The treatment effect was most remarkable in group G (30min after irradiation, PTS 12mg/mL PTS), and the skin of the mice was thickened, the elastic fibers were evenly arranged, the collagen fibers had some hyperplasia, evenly distributed, and there was no infiltration of inflammatory cells, which had significant therapeutic effect. Conclusion The total saponins of Polygala tenuifoliahas significant antioxidant activity and anti - light aging effect.
