海带提取物、枸杞子和山药混合物对糖尿病模型大鼠的降血糖作用 被引量:3

摘要 目的 研究海带提取物、枸杞子和山药混合物对链脲佐菌素诱导糖尿病模型大鼠的降血糖作用。方法 选用链脲佐菌素致高血糖SD大鼠为研究对象,用高中低三种不同剂量的海带提取物、枸杞子和山药按1:1:1配比混合,对大鼠进行灌胃治疗,阳性组给予83.5mg/kg的二甲双胍,对照组和模型组给予等量的蒸馏水,正常给药组给予高剂量药物灌胃以此来评价混合物的安全性。通过检测糖尿病模型大鼠的血糖水平、糖耐量、体质量及胰岛素变化,探究药物的降血糖机理。结果 与对照组相比,给药各组(低剂量250mg/kg、中剂量500mg/kg、高剂量1500mg/kg)的糖尿病大鼠病情均明显缓解,大鼠的毛色更为光亮,行动较为敏捷。大鼠的血糖浓度由初始的11~12mmol/L,给药30天后可降到6~8mmol/L,降血糖效果与二甲双胍组接近,胰岛素水平均有明显升高,体质量增加比率也有所上升。大鼠胰岛细胞实验表明,与空白对照组相比,给药组细胞活性增加,并呈一定的剂量依赖性。结论 海带提取物、枸杞子和山药混合物对糖尿病模型SD大鼠有明显的治疗作用,其降血糖机制可能是通过抑制胰岛β-细胞凋亡来增加胰岛素分泌。 Objective To investigate the therapeutic effect and its mechanism of pathogenicity of streptozotocin diseased rats treated by the mixture of kelp extract, wolfberry, and Chinese yam. Methods Streptozotocin-diabetic rat model were taken the mixture o f kelp extract, wolfberry and Chinese yam with the ratio of 1:1:1 by intragastric administration, positive control group was fed metformin of 83.5mg/kg, the control group and the model group were given the same amount of distilled water. The safety of high dose (1500 mg) of the mixture therapy for the normal rat were evaluated, their blood glucose levels, glucose tolerance, body weight and insulin levels were observed and its possible mechanism were analyzed. Results Compared with the model group, the disease diabetic rats of the administered group (250mg/kg, 500mg/kg, 1500mg/kg) were significantly alleviated. First of all, coat color was more shiny, the behavior were more physical agility. Followed by the decrease of blood glucose levels and AUC, and body weight and insulin levels were elevated. In cell experiments, the number of islet cells was increased in the administered group by the mixture in a dose-dependent manner. Conclusion The mixture o f kelp extract, wolfberry, and Chinese yam had obvious hypoglycemic effect. The hypoglycemic activity of the mixture may be attributed to increase insulin secretion through inhibit apoptosis of islet P-cells.
