当归多糖对辐射损伤小鼠血液中microRNA作用的研究 被引量:7

摘要 目的探讨当归多糖对60 Coγ-射线辐射损伤小鼠血液中microRNA的影响及意义。方法将35只SPF级C57BL/6J小鼠随机分为未照射组、2Gy照射未干预组、2Gy照射APS 4mg·kg^(-1)组、2Gy照射APS 8mg·kg^(-1)组、10Gy照射未干预组、10Gy照射APS 4mg·kg^(-1)组和10Gy照射APS 8mg·kg^(-1)组,共7组。照射组接受60 Coγ-射线2Gy、10Gy(剂量率为10Gy·h^(-1))全身照射后,连续3d注射APS 4mg·kg^(-1)和APS 8mg·kg^(-1),于照射后72h采集外周血。应用Agilent microRNA生物芯片筛选小鼠血液中差异表达microRNA。结果与未照射组比较,2Gy照射未干预组差异表达microRNA 30个,10Gy照射未干预组差异表达microRNA 31个,且与未照射组比较差异有统计学意义(P<0.05)。2Gy照射APS 4mg·kg^(-1)组和2Gy照射APS 8mg·kg^(-1)组之间差异表达microRNA无统计学意义(P>0.05)。10Gy照射APS 4mg·kg^(-1)组和10Gy照射APS 8mg·kg^(-1)组治疗差异表达microRNA均有所降低,除microRNA-455、microRNA-9和let-7外,10Gy照射APS 8mg·kg^(-1)组治疗降低更加显著,与10Gy照射APS 4mg·kg^(-1)组比较差异有统计学意义(P<0.05)。结论 APS通过改变血液中microRNA的表达,促进辐射小鼠的损伤修复。 Objective To ingestigate the effects of Angelica polysaccharides on the differential expression profiles of microRNA induced by 60 Coγ-ray radiation damage in mice peripheral blood and to explore their significances.Methods 35 SPF C57BL/6Jmice were randomly divived into 7groups:control group,2Gy no intervention radiation group,2Gy APS 4mg·kg^-1radiation group,2Gy APS 8mg·kg^-1 radiation group,10 Gy no intervention radiation group,10 Gy APS 4mg·kg^-1radiation group and 10 Gy APS 8mg·kg^-1 radiation group.Radiation group mice were exposed to 60 Coγ-ray,at the dosage of 2Gy,10Gy(dosage rate 10Gy·h^-1)single whole body radiation.After radiation,radiation group mice were injured with APS 4mg·kg^-1 and APS 8mg·kg^-1for 3days consecutively by injection.Peripheral blood of mice were collected after 72 h.The screening of microRNA different expression in peripheral blood was used by Agilent microRNA microarrays.Results Compared with no intervention radiation group,there was totally 30 and 31microRNA showing differential expression in 2Gy and 10 Gy no intervention group,respectively.The expression of microRNA was significant difference between control group and no intervention radiation group(P〈0.05).2Gy APS 4mg·kg^-1 radiation group and 2Gy APS 8mg·kg^-1 radiation group was no significant difference(P〉0.05).10 Gy APS4mg·kg^-1 radiation group and 10 Gy APS 8mg·kg^-1 radiation group increased the range of microRNA expression.Compared with 10 Gy APS 4mg·kg^-1radiation group,the differential expression levels of microRNA decreased obviously in 10 Gy APS 8mg·kg^-1 radiation group,excepte for microRNA-455,microRNA-9and let-7,there was statistical significance between 10 Gy APS 4mg·kg^-1radiation group and 10 Gy APS 8 mg·kg^-1 radiation group(P〈0.05).Conclusion Angelica polysaccharides may reduce microRNA differential expression in mice blood,and promote the recovery of radiation injured mice.
