稻草固体糖化发酵培养基的优化方法 被引量:1

摘要 目的探讨植物纤维素降解最佳培养基成分配比。方法以稻草粉为培养基质,接种黑曲霉进行固体发酵,以发酵产物-还原糖为标准,不同的氮源(浓度为2%的氯化铵、硝酸铵、硫酸铵、蛋白胨、酵母浸膏)、金属离子(2×10-3mol/L的Fe^(2+)、Fe^(3+)、Ca^(2+)、Na^+、Mg^(2+)、K^+)、表面活性剂(0.1%的Tween-60、Tween-80、Tween-100及SDS)进行优化配比,并通过正交设计加以验证。结果最佳发酵培养基成分为稻草粉100 g、(NH_4)_2SO_42 g、MgSO_4·7H_2O0.05 g、KH_2PO_40.1 g、Tween-80 0.1%、水200 ml、自然p H值。结论优化后的稻草固体糖化发酵培养基,可使发酵产物最大化,促进植物纤维素的开发利用。 Objective To discuss the best proportion of plant cellulose degradation medium. Methods Rice straw powder was used as culture medium,and solid fermentation was performed by Aspergillus niger. based on the fermentation product reducing sugar,different nitrogen sources( 2% ammonium chloride,ammonium nitrate,ammonium sulfate,peptone,yeast extract),metal ions( 2 × 10^-3mol / L Fe^2+,Fe^3+,Ca^2+,Na^+,Mg^2+,K^+),surfactants( 0. 1% Tween- 60,Tween- 80,Tween- 100 and SDS) were optimized. Results The best fermentation medium ingredients included 100 g straw power,2 g( NH4)2SO4,0. 05 g MgSO4·7H2O,0. 1 g KH2PO4,0. 1% Tween- 80 and 200 ml water in natural p H. Conclusion Optimized solid fermentation of solid saccharification fermentation medium for rice straw could maximize the fermented product and promote the development and utilization of plant cellulose.
