摘要
采用高效液相色谱法测定婴幼儿配方羊奶粉中酪蛋白磷酸肽(CPPs)含量。样品前处理采用超声提取,盐酸调节pH值4.4~4.6沉淀酪蛋白。流动相为0.1%甲酸水溶液和乙腈梯度洗脱。试验结果表明,在波长254 nm条件下,目标物CPPs保留时间为24.408 min。试验使用检出范围为30~300μg/mL,在此范围内CPPs呈良好的线性,线性回归方程y=375.82x+7385.4,R^2>0.9997。加标回收率在96.8%~107.3%。试验的计算标准偏差为3.3,相对偏差为0.49%。
High performance liquid chromatography was used to determine the content of CPPs in infant formula. The samples were pretreated by ultrasonic extraction, and pH was adjusted to 4.4~4,6 by hydrochloric acid to precipitate casein. The mobile phase was eluted with a gradient of 0.1% formic acid in water and acetonitrile. The experimental results showed that the retention time of CPPs was 24.408 rain at 254nm. The linear range was 30~300μg/mL, and the linear regression equation was y=375.82x+7385.4, R2〉0.9997. The recoveries ranged 96.8%~107.3%. The calculated standard deviation was 3.3 and the relative deviation was 0.49%.
