摘要
目的:以H_2O_2干预大鼠肾上腺髓质嗜铬细胞瘤细胞(PC12细胞)作为模型,评价单药楮实子水煎剂对PC12细胞凋亡的影响。方法:采用MTT法检测楮实子水煎剂对PC12细胞活性的影响,比较空白组、模型组和低、中、高浓度楮实子水煎剂在H_2O_2干预下的PC12细胞生存率。应用Western Blot法检测各组Bax、Bcl-2、细胞色素C和caspase-3凋亡相关蛋白。结果:楮实子水煎剂对PC12细胞保护作用呈浓度梯度依赖增长(P<0.05)。楮实子水煎剂能阻止H_2O_2诱导PC12细胞的凋亡,并下调cleaved-caspase-3、Bax和细胞色素C蛋白的表达,呈浓度梯度依赖(P<0.05)。结论:楮实子水煎剂对H_2O_2介导大鼠肾上腺髓质嗜铬细胞瘤细胞PC12抗凋亡的作用。
Objective: To explore the effects of Fructus Broussonetiae Decoction on the model of PC12 cells induced by H2O2. Methods: The effects of viability of PC12 cells under different concentrations of fructus broussonetiae was detected by MTI', and so was the survival rates of PCI2 ceils divided into controlled group, model group and low-dose, medial-group and high-dose group. The apoptosis-related protein such as Bax, Bcl-2, cytochrome C and caspase-3 were determined by Western Blot analysis. Results : Fructus Brousonetiae Decoction played a role in the protection of PC12 cells in a doses-dependent manner (P 〈 0. 05), and it could inhibit the apoptosis of PC12 cells induced by H2O2, and down-regulate the expression of cleaved-Caspase3, Bax, cytochrome C in a doses-dependent manner (P 〈 0. 05). Conclusion: The Frutus Broussonetiae Decotion is able to protect PC12 cells from apoptosis induced by H2O2.
