钝齿铁线莲根尖组培再生体系的研究 被引量:10

摘要 本文以钝齿铁线莲的根尖为外植体,研究不同培养基、培养条件(光培养或暗培养)以及植物生长调节剂配比对其不定芽诱导的影响。结果表明:不定芽最佳诱导培养基为添加了2.0 mg/L BA、0.5 mg/L NAA的MS+B5培养基,其不定芽诱导率高达78.5%,每个外植体平均产生8.9不定芽;最佳增殖培养基为附加有BA 2.0 mg/L和1.0 mg/L NAA改良MS培养基,增殖倍数为1.9,高度可达7.9 cm,随后转接到附加有1.0mg/L IBA,0.5 mg/L NAA和20 g/L蔗糖的改良MS培养基中,生根率达69.4%。当试管苗根形成完整根系时,把生根的试管苗移栽到装有轻型基质[V(珍珠岩:V(蛭石):V(腐殖土)=4∶1∶1]的无纺布容器中,植株成活率达到93.5%。该根尖再生体系为铁线莲大规模无性繁殖奠定了一定的基础。 Using root tip of Clematis apiifoliavar as explant, the effects of basic medium, culturing conditions, hormones and concentrations of antioxidants media on the induction of adventitious buds were investigated. The result showed that the best regeneration medium was Murashige and Skoog (MS) basal medium with Gamborg B5 vitamins supplemented with 2.0 mg/L 6-benzyladenine (BA), 0.5 mg/L naphthalenacetic acid (NAA). More than 78.5% root tip segments produced adventitious shoots, with a mean number of adventitious shoots per explant of 8.9. The optimum growing medium was observed on improved MS medium supplemented with 2. 0 mg/L BA and 1.0 mg/L NAA. Over 69.4% regeneration plantlets were able to root after transferred to the modified MS medium supplemen- ted with 1.0 mg/L indole-3-butyric acid, 0. 5 mg/L napthaleneacetic acid and 20 g/L sucrose. When micropropagated plantlets with well-developed root systems were transferred to planting bed containing a mixture of perlite, vermiculite and humus (4: 1: 1, volume ratio) in a greenhouse, 93.5%of the pla- ntlets survived. This regeneration system using root tips of C. apiifoliavar provides a foundation for large-scale vegetative propagation.
