黛黄芍药散对放射性直肠炎大鼠Th1/Th2细胞因子影响 被引量:7

摘要 目的:通过观察中药复方黛黄芍药散对放射性直肠炎大鼠肠腺存活率的影响及Th1/Th2的作用,探讨其治疗大鼠放射性直肠炎的免疫学机制。方法:将60只雌性Wistar大鼠,随机分为空白对照组、模型组、地塞米松组及黛黄芍药散高、中、低剂量组共6组,使用60Co-γ射线单次盆腔照射制作大鼠放射性直肠炎模型,在照射后第14天给药12 h后取血并处死大鼠,剥离胸腺、脾脏,称质量,计算脏器指数,取直肠组织进行HE染色,镜下观察各组肠腺存活率,采用酶联免疫吸附试验检测各组血清中干扰素-γ(Interferon-γ,IFN-γ)、白细胞介素-4(Interleckin-4,IL-4)、白细胞介素-10(Interleckin-10,IL-10)的浓度,并计算IFN-γ/IL-4。结果:较空白对照组而言,模型组肠腺存活率、脏器指数、IL-4、IL-10均出现下降(P<0.05),IFN-γ升高(P<0.01);较模型组来看,黛黄芍药散高剂量组可有效改善肠腺存活率(P<0.01)、降低IFN-γ(P<0.01)、升高IL-4(P<0.05)、平衡Th1/Th2比值(P<0.01),而黛黄芍药散中剂量组在改善脾脏指数及IL-10方面效果突出(P<0.01)。结论:黛黄芍药散可有效减轻60Co-γ射线引发的肠道损伤,抑制炎症反应,其作用机制可能与调节Th1/Th2平衡有关。 Objective : To study the effects of Daihuang Saoyao San ( DHSYS ) on crypt survival rate and Thl/Th2 in the rat model of radiation-induced proctitis produced by 60Co- γ ray, and to explore its immunological mechanism. Methods : Sixty Wistar female rats were randomly divided into 6 groups, normal group, model group, dexamethasone group, high, medium, and low dosage of DHSYS groups. Rat models of radiation-induced proctitis were produced by 60Co-γ ray radiation. The visceral indexes, crypt survival rate and serum levels of IFN- γ , IL-4, IL-10 and IFN- γ/IL-4 were detected after drug intervention for 14 days. Resuhs: Compared with the control group, the survival rate, organ index, IL-4, IL-10 in the model group were decreased ( P〈0.05 or P〈0.01 ) and the IFN- γ was increased ( P〈0.01 ). With the model group compared, DHSYS in the high dose group could effectively improve the intestinal survival rate (P〈0.01), lower IFN- γ ( P〈0.01 ), increased IL-4 ( P〈0.05 ), balance the Th1/Th2 ratio ( P〈0.01 ), while DHSYS in the middle dose group were effective in improving the spleen index and IL-10 (P〈0.01). Conclusion: DHSYS could reduce intestinal injury induced by 60Co- γ ray, inhibit inflammation, and its mechanism may be related to the regulation of Th 1/Th2 ratio.
