摘要
目的:利用响应面分析法对鬼针草叶总黄酮纯化工艺进行优化。方法:以芦丁的含量为指标,考察上样流速、上样浓度、p H值、洗脱剂浓度、柱子径高比等因素,通过Box-Behnken设计建立响应面模型来优选鬼针草叶中总黄酮的纯化工艺参数。结果:选用HPD-100型大孔吸附树脂,上样流速为2 BV/h,p H为6,上样浓度为2.027 mg/m L,70%乙醇洗脱,柱子径高比为1∶15。结论:该方法纯化工艺良好,总黄酮的纯度可达到62.86%,与理论值61.88%的相对误差为1.56%
Objective: Choosing different types of maeroporous resin to purifie total flavonoids in Bidens leaves. Methods : In the index of rutin content, study sample flow rate, sample concentration, pH, eluent concentration, column diameter to height ratio on the purification of consequences, designed to establish the response surface model to choose the purification process parameters total flavonoids in Bidens leaves and stems by Box-Behnken. Result: Adopting HPD-100 type macroporousresin, the sample flow rate is 2 BV/h, pH is 6, the sample concentration is 2.027 mg/mL, eluent concentration is 70% ethanol, the column diameter to height ratio is 1 : 15. Conclution: In this condition, the purity of total flavones could reach to 62.86%,which has relative error ( 1.56% )compared with the oretieal value ( 61.88% ).
