摘要
为观察犬肠管的组织学结构、检测犬肠管Ghrelin的表达定位情况。取3周岁母犬肠管的各段组织,通过石蜡切片制作、HE和免疫组织化学染色,对其组织学结构及Ghrelin表达定位情况进行检测。HE染色结果显示,犬肠管黏膜上皮为单层柱状上皮,黏膜肌层完整,肌层发达,肠绒毛、皱襞和肠腺等在各段肠管都存在差异。免疫组织化学检测结果显示,Ghrelin在空肠、结肠和直肠黏膜层的表达呈强阳性,阳性细胞染色为深褐色;十二指肠和盲肠黏膜层表达较弱,阳性细胞呈黄褐色;回肠未检测到Ghrelin。犬在组织学结构上和一般家畜基本相似,但黏膜肌层比较完整,肌层较厚;Ghrelin阳性细胞主要分布在犬肠管的黏膜层和黏膜下层,十二指肠、空肠、盲肠、结肠和直肠均有分布,回肠未检测到表达。
The study aims to observe the histologic structure of dog＇s intestinal canal,and to test expression and localization of Ghrelin in dog＇s intestinal canal.The intestinal canal of 3 years old female dogs was sampled,and then paraffin section,HE staining and immunohistochemical staining were conducted for detection of histologic structure and expression and localization of Ghrelin.The result of HE staining showed that the mucous membrane epithelium of the dog＇s intestinal canal was simple columnar epithelium,between intestinal villis,folds and intestinal glands.The result of immunohistochemistry testing showed that the expression of Ghrelin in the stratum mucosum was strongly positive in the jejunum,colon and rectum;weaker expression of stratum mucosum was observed in the duodenum and cecum;its experession was not detected in the ileum.The histologic structure of dog＇s are similar to general domestic animals,but more complete muscularis mucosa and more thick muscularis;Ghrelin positive cells were mainly distributed in the dog＇s intestinal stratum mucosum and submucosum,duodenum,jejunum,cecum,colon and rectum.
